摘要
目的探讨高糖对大鼠心肌成纤维细胞尾加压素Ⅱ(UⅡ)及其受体(UT)表达的影响。方法体外培养新生Wistar大鼠乳鼠的心肌成纤维细胞,随机分为正常对照组、高渗对照组、高糖组,3组细胞分别采用含5.5mmol/L葡萄糖、5.5mmol/L葡萄糖+19.5mmol/L甘露醇、25.0mmol/L葡萄糖的培养液培养48h后,应用免疫荧光法、免疫印迹法检测UⅡ及UT的表达。结果高糖组UⅡ和UT表达量明显高于正常对照组和高渗对照组,差异有显著意义(F=15.08~192.20,q=3.75~30.00,P〈0.05)。结论大鼠心肌成纤维细胞中存在UⅡ及UT表达,并且高糖刺激可使两者表达增强。
Objective To explore the effects of high glucose on the expressions of urotensinⅡ(UⅡ)and its receptor in cardiac fibroblasts. Methods The cardiac fibroblasts of neonatal Wistar rats were cultured in vitro and randomized to normal control group,hypertonic control group and high glucose group.The cells in the three groups were cultured with glucose 5.5mmol/L,glucose 5.5mmol/L+mannitol 19.5mmol/L and glucose 25.0mmol/L for 48 h,respectively.Immunofluorescence and Western bloting were used to detect the expressions of UⅡ and its receptor. Results Immumofluorescence and Western blotting analysis showed that the expressions of UⅡ and its receptor in high glucose group were higher than that in normal control and hypertonic control(F=15.08-192.20,q=3.75-30.00,P0.05). Conclusion Expressions of urotensinⅡ and its receptor exist in rat cardiac fibroblasts,and both expressions can be enhanced by high-glucose stimulation.
