摘要
目的观察依达拉奉对兔急性低氧后肠系膜血液流变学的影响。方法将50只家兔随机分为对照组、低氧模型组及低剂量依达拉奉组、中剂量依达拉奉组、高剂量依达拉奉组5组。后4组置减压氧舱内,建立急性低氧模型。在进减压氧舱前,低氧模型组和对照组经耳缘静脉给予乳酸林格液,低、中、高剂量依达拉奉组经耳缘静脉分别给予5、10及20mg/kg的依达拉奉注射液。应用微循环显微镜自动摄像分析系统观察各组出舱后兔肠系膜微循环的变化。结果低氧模型组肠系膜微循环改变符合人类急性低氧时表现。低、中、高剂量依达拉奉组微血管管径和血流速度与低氧模型组比较,差异有显著性(F=31.08~124.19,q=5.01~57.52,P〈0.05);白细胞滚动速度、沿壁滚动数、黏附数及白微栓数与低氧模型组比较,差异亦有显著意义(F=36.27~43.96,q=2.65~23.62,P〈0.05);低、中、高剂量依达拉奉组上述指标与对照组比较,差异无显著性(P〉0.05);且依达拉奉3个剂量组间上述指标比较差异无显著性(P〉0.05)。结论依达拉奉可减轻急性低氧所致的肠系膜微循环血液流变学的影响,其作用可能与直接清除自由基、降低血小板活性及抑制血小板聚集有关。
Objective To investigate the effect of Edaravone on mesenteric hemorheology in rabbits with acute hypoxia.Methods Fifty rabbits were evenly randomized to five groups as:control group;hypoxia model group;low-dose,moderate-dose,and high-dose Edaravone groups.The rats in the latter four groups were put into a low pressure oxygen cabin to create an acute hypoxia model.Before entering the low pressure oxygen cabin,the rats in the control and hypoxia model groups were given ringers solution through auricular vein,and those in low-dose,moderate-dose,and high-dose Edaravone groups were offered 5,10and20mg/kg of Edaravone,respectively,through auricular vein.A microcirculation microscope image analysis system was used to study the changes of mesenteric microcirculation. Results The changes of mesenteric microcirculation in hypoxia model group were consistent with that in human with acute hypoxia.A comparison between hypoxia model group and low-,moderate-and highdose Edaravone groups showed significant differences in terms of the microvascular diameter and blood flow velocity(F=31.08-124.19,q=5.01-57.52,P0.05),and in terms of the leucocyte rolling rate,the number of leucocyte rolling along the vessel wall,the adhesive leucocytes and microthrombus(F=36.27-43.96,q=2.65-23.62,P0.05).The differences between the three different-dose Edaravone groups and the control group were not significant with regard to the above parameters(P0.05),and the differences of the above parameters among the three different-dose Edaravone groups were not significant,too(P0.05). Conclusion Edaravone can reduce the influence on hemorheology of mesenteric microcirculation in rabbits caused by acute hypoxia.This protection may be related to directly eliminate free radicals,reduce platelet activity,and inhibit platelet aggregation.
