摘要
目的:上皮-间充质转化(EMT)能诱导 CD44+CD24-/low乳腺癌干细胞的产生,白细胞介素-6(IL-6)是很强的 EMT 诱导因子,探讨 IL-6是否通过诱导 CD44+CD24-/low细胞的产生,从而促进乳腺癌 MCF7细胞的辐射抵抗。方法含50 ng/ml IL-6的培养基培养 MCF7细胞,流式细胞术检测 CD44+CD24-/low乳腺癌干细胞标记的变化,实时定量聚合酶链反应(Real-time PCR)及Western blot 检测 EMT 标记分子的表达水平,进一步利用流式细胞术从诱导组中分选出 CD44+CD24-/low细胞,克隆形成实验检测分选后细胞的辐射抵抗能力。结果IL-6能诱导 MCF7细胞发生间充质样转化,诱导5 d 后 CD44+CD24-/low细胞显著富集,诱导形成的 CD44+CD24-/low细胞的辐射抵抗能力显著提高。结论IL-6诱导了 CD44+CD24-/low 乳腺癌干细胞的产生,从而促进MCF7细胞的辐射抵抗。
Objective To determine whether interleukin-6(IL-6),a welly established potent in-ducer of epithelial-mesenchymal transition(EMT),promotes radiation resistance of breast cancer MCF7 cellsby induction of CD44 +CD24 -/low cancer stem cells Methods Breast cancer MCF-7 cells were cul-tured in 1 0% FBS DMEMwith 50 ng/ml IL-6.The percentages of CD44 +CD24 -/low cells were measured by flow cytometry.Real-time RT-PCR and western blotting were used to determine expression of markers associated with EMT.The capacity of radiation resistance was tested by colony formation.Results IL-6 induced EMT of MCF7 cells.CD44 +CD24 -/low cells were enriched after 5-day induction of IL-6 and these induced cells exhibited enhancedradiation resistance.Conclusion IL-6 induced the generation of CD44 +CD24 -/low cells and promoted radiation resistance in breast cancer MCF7 cells.
