摘要
目的探讨卫气虚模型大鼠血淋巴细胞钟基因Clock表达的季节性变化规律。方法分别在春分、夏至、秋分、冬至前1周将20只SD大鼠随机分为正常对照组和模型组各10只,正常对照组大鼠常规饲养,模型组大鼠采用疲劳联合寒热交替法制作大鼠卫气虚证模型。1周后采用RT-PCR法检测各节气大鼠血淋巴细胞钟基因Clock表达。结果与正常对照组比较,模型组各节气钟基因Clock表达均降低(P<0.05)。与本组春分比较,正常对照组、模型组夏至、秋分、冬至钟基因Clock表达均降低(P<0.05);与本组夏至、秋分比较,正常对照组、模型组冬至钟基因Clock表达降低(P<0.05);而夏至与秋分比较,正常对照组、模型组钟基因Clock表达差异均无统计学意义(P>0.05)。结论卫气虚模型大鼠血淋巴细胞钟基因Clock表达存在春分高、冬至低的季节性节律变化,且与正常大鼠变化节律一致。
Objective To observe the seasonal change of Clock gene expression in blood lymphocytes in rats model with defensive-qi deficiency. Methods Twenty Sprague Dawley rats were randomly and equally assigned into control and model groups one week before spring equinox,summer solstice,autumnal equinox and winter solstice.Rats in control group were fed routinely,while those in model group were dealt with methods combining fatigue with alternate cold and heat stimulation to establish defensive-qi deficiency model. Clock gene expressions in blood lymphocytes in rats of both groups were measured one week later in each solar term by RT-PCR. Results Expressions of Clock gene were significantly decreased in model group than those in control group in each corresponding solar term( P〈0. 05). Compared within the same group,Clock gene expressions were significantly lower in summer solstice,autumnal equinox and winter solstice than that in spring solstice( P〈0. 05). It was significantly lower in winter solstice than those in summer solstice and autumnal equinox( P〈0. 05). However,there was no significant difference in Clock gene expression between summer solstice and autumnal equinox( P〉0. 05). Conclusion Clock gene expression in blood lymphocytes in rats with defensive-qi deficiency exhibits seasonal changes including higher in spring equinox and lower in winter solstice. These changes are in accordance with those in normal rats.
