壮骨镇痛胶囊对裸鼠乳腺癌移植瘤细胞磷脂酰肌醇3-激酶的影响

摘要 目的探讨壮骨镇痛胶囊对裸鼠乳腺癌转移瘤细胞磷脂酰肌醇3-激酶的作用。方法32只裸鼠随机等分为壮骨镇痛胶囊预防组、壮骨镇痛胶囊治疗组、参一胶囊阳性药物对照组和模型组,每组8只,接种乳腺癌细胞MDA-MB-231制备移植瘤,药物干预后,采用免疫组织化学染色法检测各组裸鼠移植瘤细胞磷脂酰肌醇3-激酶(PI3-K)的表达水平。结果与模型组比较,各药物组瘤重均显著减轻(均P<0.05),且预防组瘤重显著低于治疗组和参一胶囊阳性对照组(均P<0.05);各药物组PI3-K表达水平均显著低于模型组(均P<0.05),并且预防组PI3-K表达水平显著低于参一胶囊阳性对照组和壮骨镇痛胶囊治疗组(均P<0.05)。结论壮骨镇痛胶囊能够抑制裸鼠乳腺癌移植瘤的生长,其作用可能与抑制磷脂酰肌醇3-激酶表达水平相关,预防组较治疗组疗效更佳。 Objective To discuss zhuanggu zhentong capsule on the role of phosphatidyl inositol 3-kinase of nude mouse breast cancer metastatic tumor cells. Methods 32 nude mice were randomly divided into four groups,named as the prevention group of zhuanggu zhentong capsule; the treatment group of zhuanggu zhentong capsule; the positive drug control group of shenyi capsule and the blank model group. Each group was consisted of eight nude mice. After the nude mice implanted breast cancer cells and drug intervention,we would calculate and contrast the inhibition rate of tumor and phosphatidyl inositol 3-kinase of tumor cells for each group of nude mice. Results Among the positive drug control group,the prevention group and the treatment,it was found that the inhibition to the growth and the expression of PI3-K in human breast cancer cells in nude mice transplanted tumor. But the effect of the prevention group of zhuanggu zhentong capsule was more apparent. Conclusions Zhuanggu zhentong capsule can restrain the activity of phosphatidyl inositol 3-kinase expressed in nude mouse.The effect of the prevention group is better than that of the treatment group.
