摘要 为建立高效液相色谱-蒸发光散射检测法(HPLC-ELSD)同步定量测定烟草中果糖、葡萄糖的新方法,将烟草样品处理后,用水提取,用Kromasil NH2柱分离,以乙腈-水(80∶20,V/V)为流动相,流速1.0 m L/min,柱温为室温,进样量20μL,ELSD漂移管温度95℃,空气作载气,流速1.5 L/min,使用蒸发光散射检测器进行了单糖检测.结果表明:果糖、葡萄糖的线性范围分别为0.273~2.73 mg/m L,0.294~2.94 mg/m L,在线性范围内糖含量与峰面积线性关系良好;样品中糖的加标回收率范围为98.5%~102.8%,测得烟草样品中果糖、葡萄糖含量分别为10.44%、7.40%故该方法具有快速、有效、质量可控等优点,为烟草中糖含量的测定提供了一种行之有效的分析方法. A new evaporative light scattering detection( ELSD) based HPLC method( HPLC-ELSD) was established for the simultaneous quantitative analysis of fructose and glucose in tobacco. The treated tobacco samples were extracted with water and separated on Kromasil NH2 column by using acetonitrile and water( 80 ︰ 20,V / V) as the mobile phase at a flow rate of 1. 0 m L / min,with a sample size of 20 μL and the column temperature at room temperature. The ELSD drift tube temperature was set at 95℃ and air was used as the carrying gas( 1. 5 L / min). The results indicated that the linear ranges of fructose and glucose were 0. 273 ~ 2. 73 mg / m L and 0. 294 ~ 2. 94 mg / m L,respectively,and the sugar content had good linear relationship with the peak area in the linear range. The sugar recoveries ranged from 98. 5% to 102. 8% for the control experiment. The contents of fructose and glucose in tobacco were determined as 10. 44%,7. 40% respectively.It was proved to be a rapid,effective,and controllable method for the determination of monosaccarides in tobacco.
