EBV编码miRNA的研究进展

摘要 MiRNA(microRNA,miRNA)是一组长约21~25个核苷酸的内源性的非编码的小RNA。miRNA最初在细胞核中表达,即初级RNA(primary micro RNA,pri-miRNA),pri-miRNA在核酸酶Drosha RNase的作用下,剪切为具有发夹结构的前体microRNA(precursor micro RNA,pre-miRNA),由输出蛋白5转运到细胞质中,在核酸酶Dicer和辅助因子TRBP的作用下,Pre-miRNA被剪切成21~25个核苷酸长度的双链miRNA,双链解螺旋,一条链立即降解,而另一条链形成RNA诱导的沉默复合体(RNAinduced silencing complex,RISC),若RISC复合物中的miRNA与靶信使RNA(messenger RNA,mRNA)的3’端非编码区不完全互补配对时,抑制靶mRNA转录,若为完全互补配对时,则降解靶mRNA,从而调节基因的表达”。
