摘要
针对中国首例实验室确诊输入性MERS-CoV感染病例,采用鼻咽拭子样品进行核酸提取、基因扩增与测序,获得MERS-CoV_ChinaGD01结构蛋白与附属蛋白编码基因,包括S基因、E基因、M基因、N基因、ORF3、ORF4a、ORF4b、ORF5和ORF8b基因序列。根据S基因进化分析发现中国输入性病例中MERS-CoV虽有少数位点变异,但仍与近年沙特流行株相近,属于MERS-CoV亚型5,N、E、M等结构基因核苷酸变异较少。附属蛋白进化分析表明:ORF3、ORF4a、ORF4b、ORF5均有一定的核苷酸替换,而ORF8b相对保守。总之,中国首例输入性MERSCoV与已发表序列相比总体表现为保守,但在部分基因序列上有一定的变异。这是中国首例输入性MERS-CoV的第一次基因分析结果报道,可为该MERS-CoV感染特点的进一步研究及相关疾病的防控提供参考。
To investigate the genetic character and origin of the first imported infection case of middle East respiratory syndrome coronavirus(named as MERS-CoV_China GD01),RNA was extracted from swabs of this patient followed by RT-PCR amplification.All coding gene of structural(S,E,M,E)and accessory(ORF3,ORF4 a,ORF4b,ORF5,ORF8b)proteins were sequenced and analyzed.Phylogenetic analyses of structural protein coding genes of MERS-CoV_China GD01 indicates that several substitutes exists in S coding gene and its origin belong group 5of MERS-CoV,which were recent circulated in Saudi Arabia area,while other three structural genes(N,E,M)were very conserved.Phylogenetic analyses of accessory protein coding genes of MERS-CoV_China GD01 indicates that several substitutes exists among ORF3,ORF4 a,ORF4band ORF5,while ORF8 b was conserved.In conclusion,genome of MERS-CoV_China GD01 was general conserved although several genetic variations were found among structural and accessory protein coding genes.This is the first report on sequencing and phylogenetic analyses of the first imported MERS case in China,which may pay the way for prevention and control of imported MERS-CoV infection.
