血啉甲醚对蜡样芽孢杆菌孢子的光动力杀伤作用

摘要 目的探讨血啉甲醚(HMME)对蜡样芽孢杆菌孢子的杀伤作用。方法通过蜡样芽孢杆菌孢子失活率测定以及荧光显微镜和原子力显微镜图像观察HMME对蜡样芽孢杆菌孢子的光动力杀伤作用。结果一定范围内,HMME浓度越大,对孢子的杀伤作用越强;荧光显微镜观察加了HMME的孢子能发出荧光,而且浓度越高,发荧光的孢子数量越多;原子力显微镜观察蜡样芽胞杆菌孢子加HMME后形态基本没有变化。结论 HMME通过进入孢子内部导致其异常生理变化,引起内容物如蛋白质、核酸等的氧化损伤而达到杀伤作用。 Objective To study photodynamic inactivation of hematoporyrin monomethyl ether(HMME) to Bacillus cereus spores.Methods The reaction was discussed by inactivation rate and the observations of fluorescence microscopy and atomic force microscope(AFM).Results The killing effect was enhanced with the HMME concentration increasing at certain range; the spores added with HMME could fluoresce and the number of fluorescent spores was growing with the HMME concentration increasing by fluorescence microscopy obervation,however from the images of AFM,the shapes of spores added with HMME were unchanged.Conclusion The physiological changes of spores occurred abnormally by the entering of HMME into its body,causing the oxidative damage of the spores ' contents such as protein and nucleic acids to achieve the killing effect.
