IL-22及IL-22siRNA对HaCaT细胞分泌HB-EGF的影响

Effects of IL-22 and IL-22 siRNA on the Expression of Heparin-binding Epidermal-growth-factor-like Growth Factor in HaCaT Cells

摘要 目的研究IL-22及IL-22siRNA对HaCaT细胞肝素结合表皮生长因子样生长因子(heparin—bindingepidermalgrowthfactor-1ikegrowthfactor,HB—EGF)表达的影响。方法用12.5~100μg/L的IL-22和IL-22siRNA干预处理HacaT细胞,继续培养24h后,分别用实时荧光定量RT—PCR和Westernblot法、ELISA法检测IL-22及IL-22siRNA对HaCaT细胞HB—EGFmRNA和蛋白的影响。结果不同浓度的IL-22干预处理后,HB-EGF的mRNA和蛋白水平均较对照组升高,且随着IL-22浓度的升高,其促HB-EGF的表达作用逐渐增强,与对照组相比,差异具有统计学意义(P〈0.05)。IL-22siRNA处理组的HB-EGFmRNA和蛋白水平较对照组低,与对照组相比,差异具有统计学意义(P〈0.05)。结论IL-22可剂量依赖促进HB-EGF的表达,IL-22siRNA可抑制HB-EGF的表达。 Objective To investigate the effects of interleukin-22 (IL-22) and IL-22 siRNA on HB-EGF expression in HaCaT cells. Methods After being treated with 12. 5 - 100μg/L IL-22 and IL-22 siRNA for 2gh,total RNA and total protein were collected. Level of HB-EGF mRNA was examined by real-time quantitative RT-PCR, and protein level of HB-EGF was detected by western blot and ELISA. Results Compared with the control, both mRNA and protein level of HB-EGF were elevated with the increase concentration of IL-22 treatment, and the promoting function gradually enhanced. However, the expression of HB-EGF decreased significantly after IL-22 siRNA exposure. The difference were statistically significant in IL-22 and IL-22 siR- NA. Conclusion IL-22 treatment can dose-dependently induce HB-EGF expression, IL-22 siRNA can in- hibit HB-EGF expression.
