The Influence of PEEK/HA Bio-composites on 3T3 Fibroblasts

摘要 体外研究PEEK/HA生物复合材料对3T3成纤维细胞的影响,评价该材料的生物相容性。制备不同HA含量的PEEK/HA和PEEK/SPEEK/HA生物复合材料,与3T3成纤维细胞共培养,倒置相差显微镜观察该材料对细胞形态影响,MTT法研究该材料对细胞增殖作用,评价细胞毒性,流式细胞术研究该材料对细胞凋亡的影响,扫描电镜观察该细胞在材料表面黏附形态。细胞呈长梭形贴壁生长,胞体透明,大部分呈梭形和多角形,生长态势良好。随着浸提液浓度增大,材料对细胞增殖作用显著下降,PEEK/HA复合材料随着HA含量增加,细胞增殖作用逐渐增强,PEEK/SPEEK/HA复合材料与PEEK/HA相比,对细胞增殖作用较强,细胞毒性级别为Ι级,说明该材料对3T3细胞无毒性。细胞凋亡率随PEEK/SPEEK/HA材料中的HA含量升高而降低。SPEEK的加入使PEEK/HA复合材料后黏附率增大,随PEEK/SPEEK/HA中HA含量增加,细胞黏附率增加,SPEEK和HA可改善材料的黏附性能。SEM观察发现3T3细胞能够在PEEK/SPEEK/HA复合材料表面黏附、伸展和生长。PEEK/SPEEK/HA复合材料生物相容性良好,为该人工骨替代材料的临床前研究提供实验数据和科学依据。 The PEEK/SPEEK/HA composites and PEEK/HA composites with different HA contents were prepared, and the fibroblasts were cultured with the composite materials. The influences of PEEK/HA bio-composites on 3T3 fibroblasts in vitro were researched, and biocompatibility of the materials was evaluated. The cell morphology was observed by inverted fluorescence microscope. The MTT method was used to study the cell proliferation under various conditions to evaluated cytotoxicity. The effect of this material on the apoptosis was studied by flow cytometry, and the ceils adherent morphology on the material surface was observed by SEM. With the increasing of concentration of the composites extracts, the cell proliferation was decreased, and with increasing the HA in PEEK/HA material, the cell proliferation was enhanced. Compared with PEEK/HA material, the PEEK/SPEEK/HA had a greater effect on cell proliferation and with the increasing of HA content, PEEK/SPEEK/HA materials could accelerate cell proliferation. The cytotoxicity of PEEK/SPEEK/HA composite and PEEK/HA composite was Ⅰ level, it indicated that the materials were non-cytotoxic. With the increasing of HA content in PEEK/SPEEK/HA materials, the apoptosis rate reduced. After the SPEEK was added to PEEK/HA composite, the adhesion rate increased, with the increasing of HA content in PEEK/SPEEK/HA, the cell adhesion rate also increased, it showed that the SPEEK and HA could improve the adhesion properties of the materials. 3T3 cells could adhere, stretch and grow on the surface of PEEK/SPEEK/HA composites, and the PEEK/SPEEK/HA composite materials had good biocompatibility, and this research would provide experimental data and scientific basis for pre-clinical studies on artificial bone substitute materials.
