摘要
600只7日龄海兰褐蛋雏鸡随机分为10个处理,每个处理60只,分为3个重复,每个重复20只。其中5个处理为免疫抑制组,每天灌服环磷酰胺(CY)100mg/kg,其余为正常饲喂组,灌服等量生理盐水,连续7d。免疫抑制组和正常饲喂组基础饲粮中人参茎叶多糖(Ginseng stem and leaf polysaccharide,GSLP)添加干物质量同为0.2、0.4、0.8、1.6g/kg。雏鸡35日龄时,每个处理中随机选择9只雏鸡(每个重复3只),采集十二指肠、空肠和回肠样品,利用比色法、ELISA和real-time PCR方法分析一氧化氮(NO)、白细胞介素-2(IL-2)和分泌型免疫球蛋白A(slgA)含量以及干扰素-γ(IFN-γ)mRNA表达水平。结果显示,1.6g/kg GSLP显著提高了空肠IL-2含量(P〈0.05),并且IL-2含量与GSLP呈正相关关系(P〈0.10);环磷酰胺则显著降低了空肠IL-2含量(P〈0.01);GSLP对其它指标无显著影响。结果表明,0.2-1.6g/kg添加范围内,不同免疫状态雏鸡空肠黏膜细胞免疫水平随GSLP添加量的增加而提高;灌服CY对肠道黏膜细胞免疫的抑制的后续影响可以持续到35日龄。
The objective of this study was to observe the effects of ginseng stem and leaf polysaccharide(GSLP)on cytokine and immunity mediator secretion of chicken small intestinal mucous tissue with different immune states.Six hundred(7days of age)healthy Hy-line variety brown chicken(52.79±4.58)g were freely divided into 10 treatments,60chicken per treatment with 3replications of 20 chicken each,in which,5treatments were chosen as immune depressed group and drenched with 100mg/kg cyclophosphamide(CY),the others were treated as normal feeding with the same amounts of physiological saline,continued for 7days.GSLP levels of two groups were 0,0.2,0.4,0.8and 1.6g/kg in each group based on basal diet,respectively.At 35 d,9chicken of each treatment(3per replication)were freely selected for collection of duodenum,jejunum and ileum samples.The methods of colorimetry,ELISA and real-time PCR were utilized for analyzing the contents of nitric oxide(NO),interleukin-2(IL-2)and secreted IgA(sIgA),and mRNA expression of interferon-γ(IFN-γ).The results showed that,1.6g/kg GSLP significantly improved the level of IL-2in jejunum(P〈0.05),being notable positive correlation with GSLP(P〈0.10);CY significantly decreased IL-2content in jejunum(P〈0.01);but GSLP had no obvious effects on other indexes.In conclusion,jejuna mucosa cellular immunity tends to be improved by GSLP additive(from 0.2g/kg to 1.6g/kg);the subsequent depressed effects of drenched CY on intestinal mucous cellular immunity can be observed in 35 days of age.
