摘要
探讨玉米赤霉烯酮(Zearalenone,ZEA)对雌性大鼠子宫内膜细胞凋亡与Caspase-3和Caspase-8蛋白表达的关系,进而揭示其导致子宫损伤的作用机制。应用HE染色观察组织病理变化,免疫组化SP法检测注射玉米赤霉烯酮后6个时间点大鼠子宫内膜细胞Caspase-3和Caspase-8蛋白的表达。病理组织学观察发现,试验组子宫组织出现不同程度损伤,子宫内膜细胞发生凋亡。免疫组化结果显示,试验组子宫内膜上皮细胞与间质细胞Caspase-3和Caspase-8蛋白表达量均明显高于对照组(P<0.05)。不同时间点相比,Caspase-3在内膜上皮细胞中12h表达量最高,而在内膜间质细胞中18h表达量最高;Caspase8在内膜上皮细胞中18h表达量达到峰值,而在内膜间质细胞中24h表达量最高,之后缓慢下降。结果表明,ZEA可能通过上调Casepase-3和Caspase-8的表达从而引起子宫内膜细胞凋亡。
This study aimed to investigate the relationship between endometrial cells apoptosis in rats exposured to zearalenone(ZEA)and the expression of Caspase-3and Caspase-8,and then revealed the mechanism that cause uterus damage.HE stain was used to observe the pathological changes and immunohistochemistry SP method to detect the expression of Caspase-3and Caspase-8in endometrial cells after injection of ZEA(corn oil)at six time points.Pathologichistology elucidated the injuried uterus tissue in different degree and endometrial cells apoptosis.The results of immunohistochemistry indicated that the expression of Caspase-3and Caspase-8in trial group were significantly higher than that of control group(P〈0.05).The highest peak of Caspase-3in endometrial epithelial cells emerged at 12 h,and that in endometrial stroma cells emerged at 18 h.The expression of Caspase8 in endometrial epithelial cells increased to peak value at 18 h,and that in endometrial stroma cells reached to the highest at 24 hand then slowly decreased.The results showed that ZEA induced apoptosis of endometrial cells that might be through up-regulating the expression of Caspase-3and Caspase-8.
