摘要
用含不同浓度(0、0.04%、0.08%、0.12%)水蛭素的稀释液稀释美系长白公猪精液,0.5mL细管分装冷冻,分别检测平衡后及冻融后精液中过氧化氢酶的活性和mRNA表达量的变化。结果显示,不含水蛭素的冷冻稀释液显著降低新鲜猪精液过氧化氢酶活性(P<0.05),而含有0.08%水蛭素的冷冻稀释液显著提高冷冻保存猪精液过氧化氢酶活性(P<0.05);不含水蛭素的冷冻稀释液降低新鲜猪精液过氧化氢酶mRNA的表达,含有0.08%水蛭素的猪精液平衡及冻融后的过氧化氢酶mRNA表达量最高,但差异不显著(P>0.05)。结果表明,添加0.08%水蛭素显著提高冷冻前后猪精液过氧化氢酶活性(P<0.05),但对过氧化氢酶mRNA表达的影响并不显著。
The purpose of the study was to evaluate how the hirudin affect the enzyme activity and the mRNA expression of catalase in boar semen.during cryopreservation and improve the quality of cryopreserved boar semen.Semen collected from American-derived landrace boars was diluted with extenders containing different concentrations(0,0.04%,0.08% and 0.12%)of hirudin and frozen in 0.5 mL straws,measured catalase enzyme activity and the expression of mRNA from different phases of cryopreservation.The results showed that the extenders without hirudin had significantly negative effect on the activity of catalase enzyme in fresh boar semen(P〈0.05),but extenders with 0.08% hirudin has positive effect on the activity of catalase enzyme in boar semen during cryopreservation(P〈0.05);extenders without hirudin reduced the expression of catalase mRNA in fresh boar semen(P〉0.05),but the expression of catalase enzyme mRNA in boar semen with 0.08% hirudin during cryopreservation was the highest(P〉0.05).In brief,extenders with 0.08% hirudin has significantly positive effect on the activity of catalase enzyme in boar semen during cryopreservation,but the effect on the expression of catalase mRNA is not significant.
