摘要
建立了一种鉴定生物活性多肽的方法,该方法通过计算机程序将MS分析得到的分子量与已知牛乳蛋白质氨基酸序列比对,得到预测的多肽序列,再根据该序列二级质谱图的az、by断裂情况,经过Biolynx软件分析证实是否为该序列。通过这种方法得到了来源于牛奶蛋白的119条多肽序列,包括已被前人报道过的具有ACE抑制活性的SG,LLP,QSLVY,VDGKEDL,有抗菌活性的EKF和KKISQ等。此外,还有些未被报道过的多肽序列。该方法能普遍用于复杂基质中生物活性肽的分析鉴定。
This research establishes a method to identify peptide, whichget possible sequence by matching molecular weight obtained with milk protein database, then verifies sequence by Biolynx. 118 peptides from milk proteins are identified, including SG、 LLP, QSLVY,VDG- KEDL which were reported as ACE inhibitor, EKF, KKISQ as antimicrobial peptides etc. Moreover, this method has also identified some novel peptides. This method can be used for identifying bioactive peptides in complex matrices, such as fermented milk.
