泛素蛋白酶体通路与动脉粥样硬化

摘要 泛素-蛋白酶体通路(ubiquitin-proteasome pathway,UPP)是广泛存在于真核细胞中的,依赖ATP的选择性蛋白质降解的主要途径。泛素的主要功能是标记细胞中需要降解的蛋白质,使其被26S蛋白酶体识别和降解。UPP主要通过介导细胞中蛋白的降解而发挥调节作用。该通路的异常与许多疾病诸如肿瘤、遗传性疾病、神经退行性疾病、心血管疾病等密切相关。本文主要综述了UPP在动脉粥样硬化的发生,发展和复合病变中的作用。 Ubiquitin - proteasome pathway (UPP) is the major selective ATP-dependent protein degradation path- way which is widely in eukaryotic cells. Substrates are firstly targeted by covalent attachment of multiubiquitin chains, then recognited and degradated by 26S proteasome. UPP takes the regulation roles via UPP-mediated protein degradation. UPP is involved in a number of diseases such as cancer, genetic diseases, neurodegenerative diseases, cardiovascular diseases and so on. In this review, the role of UPP in the occurrence, development and complex lesion of arteriosclerosis (AS) will be summarized.
