5-氨基酮戊酸诱导白念珠菌生成原卟啉Ⅸ两种检测方法的比较

摘要 目的对两种测定原卟啉Ⅸ(ProtoporphyrinⅨ,PpⅨ)荧光强度的方法进行比较,探求高效、准确、可靠、快速的检测方法。方法将白念珠菌悬液与5-氨基酮戊酸(5-Aminolevulinic acid,5-ALA)避光孵育。分别利用荧光分光光度计(以下简称荧光法)和共聚焦显微镜(以下简称共聚焦法)下测量PpⅨ荧光强度。结果两种测量方法的统计学比较P<0.05,差异有显著性。共聚焦显微镜操作简单,方便快捷,自带分析软件可以直接算出测值,计算精确,但是测量价格较贵,荧光分光光度计相对操作相对繁琐,但是价格便宜。结论共聚焦显微镜无论从测量的简便性还是从测量的准确性都要优于荧光分光光度计。
