摘要
目的观察灯盏花素对大鼠缺血再灌注后脑组织白细胞介素(IL)-1β、IL-10、IL-18和肿瘤坏死因子-α(TNF-α)表达的影响。方法选用健康雄性SD大鼠48只,随机分为4组:假手术组、模型组、川芎嗪组和灯盏花素组。制作大鼠大脑中动脉脑缺血再灌注模型,分别于血流阻断1小时后,缺血再灌注24、48、72小时后,断头取脑,采用酶联免疫吸附测定(ELISA)检测大鼠脑组织中IL-1β、IL-10、IL-18和TNF-α的表达水平。结果川芎嗪和灯盏花素均可显著降低大鼠脑皮层区和海马区IL-1β、IL-18和TNF-α的含量,同时升高大鼠脑皮层区和海马区IL-10的含量。且同川芎嗪相比,灯盏花素可显著降低大鼠脑皮层区和海马区IL-1β、IL-18和TNF-α的含量,升高大鼠脑皮层区和海马区IL-10的含量。结论灯盏花素可通过降低大鼠脑皮层区和海马区IL-1β、IL-18和TNF-α的含量,并升高大鼠脑皮层区和海马区IL-10的含量,发挥神经保护作用。
Objective To observe effects of breviscapine on the expression of interleukin(IL)-1β, IL-10, IL-18 and tumor necrosis factor-α(TNF-α) in rats with focal cerebral ischemia reperfusion. Method 48 rats were randomly divided into four groups: sham operation group, focal cerebral ischemia-reperfusion group, tetramethylpyrazine group and breviscapine group. The middle cerebral artery occlusion reperfusion model were made by the suture method, after ischemia reperfusion 24, 48 and 72 hours, the rats were killed and gained the brain. The level of IL-1β, IL-10, IL-18 and TNF-α were measured by enzyme linked immunosorbent assay(ELISA). Result Tetramethylpyrazine and breviscapine could significantly decreased the level of IL-1β, IL-18 and TNF-α in rat's cerebral cortex and hippocampus, and increase the level of IL-10 in rat's cerebral cortex and hippocampus. Compared with tetramethylpyrazine, breviscapine significantly decreased the level of IL-1β, IL-18 and TNF-α in rat's cerebral cortex and hippocampus, and increased the level of IL-10 in rat's cerebral cortex and hippocampus. Conclusion Through decrease the level of IL-1β, IL-18, TNF-α and increase the level of IL-10 in rat's cerebral cortex and hippocampus, breviscapine plays a neuroprotective effect in rats.
