摘要
为提高长角血蜱副肌球蛋白(Pmy)的表达量,本研究对所在课题组前期构建的含有p ET-32(a+)-pmy的E.coli BL21载体的表达条件进行优化,考察温度、时间和异丙基硫代-β-D-半乳糖苷(IPTG)浓度对Pmy表达量的影响.SDS-PAGE电泳结果表明,最优表达条件为:诱导温度37℃,诱导时间8 h,IPTG浓度0.4 mmol/L.
To improve expression level of paramyosin(Pmy)from Haemaphysalis longicornis,the expression conditions of E. coli BL21 recombined with p ET-32(a+)-pmy before in our laboratory were optimized. Three factors were involved in this study,including tempera-ture,time and concentration of isopropyl β-D-1-thiogalactopyranoside(IPTG). The results of SDS-PAGE showed that the op timal temperature was 37 ℃,the inducing time was 8 h,and the concentration of IPTG was0.4 mmol /L.
