G蛋白耦联受体5通过上皮-间充质转化促进结肠肿瘤侵袭转移的临床研究 被引量:5

摘要 目的分析G蛋白耦联受体5(Lgr5)在结肠癌侵袭转移过程中的作用及其意义。方法采用免疫组织化学法检测97例结肠癌组织标本及其对应正常结肠组织标本中Lgr5、E-cadherin、N-cadherin的表达情况。结果在结肠癌中,Lgr5在肿瘤组织中表达阳性率为55.7%(54/97),正常组织中Lgr5表达阳性率为5.2%(5/97)。Lgr5在肿瘤组织与正常组织中表达比较差异有统计学意义(χ^2=54.480,P〈0.05)。E—cadherin在肿瘤组织中表达阳性率为51.5%(50/97),正常组织中E-cadherin表达阳性率为87.6%(85/97)。E—cadherin在肿瘤组织与正常组织中表达差异具有统计学意义(χ^2=29.837,P〈0.05)。N—cadherin在肿瘤组织中表达阳性率为66%(64/97),正常组织中N-cadherin表达阳性率为15.5%(15/97),N—cadherin在肿瘤组织与正常组织中表达比较差异有统计学意义(χ^2=51.271,P〈0.05)。Lgr5的表达,结肠癌淋巴结有无转移比较差异有统计学意义(χ^2=11.178,P〈0.05),有无远处转移比较,差异有统计学意义(χ^2=7.900,P〈0.05)。Lgr5的表达和E-cadherin的表达呈负相关(r=-0.562,P〈0.05),和N-cadherin的表达呈正相关(r=0.458,P〈0.05)。结论Lgr5表达水平的改变能够影响结肠癌细胞的侵袭转移能力,Lgr5可能促进结肠肿瘤细胞EMT的发生,参与了结肠癌的侵袭转移。 Objective To study the roles and significance of Lgr5 in invasion and metastasis of colon cancer. Methods Immunohistochemisruy was employed to detect the expression of Lgr5, E-cadherin and N-cadherin in 97 samples of colon cancer tissues and normal tissues. Results The positive rate of Lgr5 expression was 55.7% (54/97) in colon cancer tissues and 5.2% (5/97) in normal tissues, and there was significant difference between groups ( χ^2 = 54. 480, P 〈 0.05 ). The positive rate of E-cadherin ex- pression was 51.5% (50/97)in colon cancer tissues and 87.6% (85/97)in normal tissues, and there was significant difference between groups( χ^2=29. 837, P 〈 0.05 ). The positive rate of N-cadherin expression was 66% (64/97)in colon cancer tissues and 15.5 % (15/97)in normal tissues, and there was significant difference between groups( χ^2 = 51. 271 ,P 〈 0. 05 ). In colon cancer tissues, the expression of Lgr5 was correlated with lymph node metastasis ( χ^2 = 11. 178, P 〈 0. 05 ) and distant metastasis ( χ^2 = 7.900, P 〈0. 05 ). The expression of Lgr5 protein was negatively correlated ( r = - 0.562, P 〈 0. 05 ) to the E-cadherin protein, but it was positively correlated to the N-cadherin protein ( r = 0. 458, P 〈 0.05 ). Conclusion Lgr5 may participate in the occurrence of EMT, affect the expression of E -cadherin and N-cadherin, and may be related to the metastasis and progression of colon cancer.
