有氧运动对非酒精性脂肪肝大鼠肝组织AMPK蛋白活性的影响 被引量:12

摘要 目的:观察有氧运动对非酒精性脂肪肝大鼠肝组织AMPK蛋白活性的影响,并探讨其作用机制。方法:30只SD雄性大鼠随机分为正常对照组(N组)、脂肪肝模型组(C组)和运动组(E组),N组喂食基础饲料,C组和E组均喂食高脂饲料,E组大鼠进行12周游泳训练。12周末取材比较各组大鼠体重、肝脏湿重及摄入总热量。观察肝组织切片,并对其炎症活动度评分。检测比较各组大鼠肝组织中TC、TG、FFA含量和血清TC、TG、ALT、AST、FFA含量变化;肝组织AMPK蛋白的活化水平。结果:12周实验结束后,C组大鼠和E组大鼠摄入总热量无显著性差异,但显著高于N组。C组大鼠体重及肝脏湿重显著高于N组和E组。肝组织切片显示,N组大鼠肝组织学特征正常,细胞形态正常,肝板排列整齐,没有肝细胞脂肪变性及炎症浸润;C组大鼠肝组织可见弥漫性肝细胞脂肪变性,肝板排列不整齐,肝小叶内炎症细胞浸润,出现汇管区炎症;E组大鼠肝细胞形态基本正常,肝板排列较整齐,肝小叶内及汇管区少见炎症细胞浸润。C组大鼠肝脏炎症活动度评分显著高于N组和E组。C组大鼠肝组织TC、TG、FFA水平显著高于N组和E组。C组大鼠血清TC、TG、FFA、ALT、AST水平均显著高于N组和E组。C组大鼠肝组织p-AMPK和p-AMPK/AMPK水平显著低于N组和E组。结论:有氧运动通过激活并提高肝组织p-AMPK蛋白表达水平改善非酒精性脂肪肝大鼠的脂质代谢,从而发挥防治非酒精性脂肪肝的作用。AMPK是有氧运动防治非酒精性脂肪肝的重要作用靶点。 Objective This experiment was to investigate the effect of aerobic exercise on the AMPK protein activity in nonalcoholic fatty liver. Methods 30 male SD rats were randomly assigned to control group(N),high fat diet group(C),high fat diet and exercise group(E). Rats in the group E were forced to swim for 12 weeks.By the end of the experiment,the expression of AMPK in liver tissue were tested and the contents of TC,TG,and FFA in liver tissue and TG,TC,FFA,ALT,and AST in serum of the rats were detected. Results After 12 weeks of experiment,the body weight and liver wet weight in group C were obviously higher than the other two groups. The serum TG,TC,ALT,and AST in group C was higher than that in the other two groups. The highest contents of TC,TG,and FFA in liver tissues in group C were observed. The expression of p-AMPK and p-AMPK/AMPK in liver of groups N and E were significantly higher than that of group C. Conclusion Results suggested that aerobic exercise could increase p-AMPK and p-AMPK/AMPK in liver tissue,improve the lipid metabolism,and thus prevent the occurrence of high fat diet-induced nonalcoholic fatty liver in rats.
