急性淋巴细胞白血病患者Notch1和ASB2基因mRNA表达的研究

摘要 目的:研究急性淋巴细胞白血病(acute lymphoblastic leukemia,ALL)患者骨髓中单个核细胞Notch1和ASB2基因mRNA的表达,探讨急性淋巴细胞白血病中Notch信号通路与蛋白泛素化作用的相关性,为白血病发生发展的分子机制和临床治疗提供有价值的参考。方法:收集初诊的48例ALL患者以及34例非白血病和健康人(对照组)骨髓,采用实时荧光定量PCR检测骨髓中Notch1和ASB2 mRNA表达情况。48例ALL患者中,其中B细胞ALL 37例,T细胞ALL 11例。结果:B-ALL和T-ALL患者骨髓中Notch1 mRNA表达量相对于对照组分别升高了630.3倍和4608倍,差异均有统计学意义(t=42.9,46.7,P<0.01),并且T-ALL患者骨髓中Notch1 mRNA表达量明显高于B-ALL,差异有统计学意义(t=8.66,P<0.01)。B-ALL和T-ALL患者骨髓中ASB2 mRNA表达量均高于对照组(t=24.126,17.82,P<0.01)。Notch1与ASB2 mRNA表达量具有相关性(r=0.468,P<0.01)。结论:ALL患者骨髓存在Notch1-ASB2模式的异常表达,该机制可能与白血病的发生发展有关。 Objective: To explore the mRNA expression of Notch1 and ASB2 mRNA in bone marrow plasma mononuclear cells of patients with acute lymphoblastic leukemia and discuss the correlation between Notch1 signaling pathway and ubiquitination in acute lymphoblastic leukemia,which may provide valuable information for investigating the pathogenesis of acute lymphoblastic leukemia and clinical treatment. Methods: Bone marrow plasma was collected from 48 patients of newly diagnosed acute lymphoblastic leukemia( 37 cases B-ALL and 11 cases T-ALL) and 34non-hematopathy and healthy individuals( control group). Expression levels of Notch1 and ASB2 mRNA were detected by real-time quantitative PCR. Results: The expression of Notch1 mRNA in B-ALL and T-ALL were significant different compared to healthy individuals group( t = 42. 9,46. 7,P〈 0. 01),which were 630. 3,4608 folded more than healthy individuals,and the expression of Notch1 mRNA in T-ALL was significantly increased than in B-ALL( t = 8. 66,P〈 0. 01). Morever,the expression of ASB2 mRNA in B-ALL group and T-ALL group were significantly increased than healthy individuals( t = 24. 126,17. 82,P〈 0. 01). The expression of Notch1 and ASB2 gene was positive correlaed( r = 0. 468,P〈 0. 01). Conclusion: An abnormal Notch1-ASB2 expression was found in patients with ALL,which may be related to leukemogenesis.
