摘要
目的研究自发性高血压大鼠(SHR)脑组织核转录因子(NF-κBp65)与凋亡调节因子Bcl-2和Bax表达的变化,并探讨星状神经节阻滞对这一过程的影响。方法将32只10周龄雄性SHR随机分为四组,每组8只,即左侧星状神经节阻滞组(LS组)、右侧星状节阻滞组(RS组)、药物卡托普利组(D组)、手术对照组(C组)。实验结束后用3%戊巴比妥钠腹腔注射(45 mg/kg)麻醉自发性高血压大鼠,迅速取出脑组织,免疫组化检测脑组织Bcl-2、Bax表达,放射免疫法测定脑组织NF-κBp65的含量。结果 RS组和D组脑组织NF-κBp65的含量显著低于LS组和C组(P<0.05),而LS组和C组,RS组和D组比较差异未见统计学意义(P>0.05);与LS、D、C组相比,RS组SHR脑组织Bcl-2表达明显增强,Bax表达明显减弱,Bcl-2/Bax比值明显增高(P<0.05)。结论右侧星状神经节阻滞可通过影响脑内NF-κBp65和凋亡调节基因的表达而在脑组织保护中发挥一定的作用。
Objective To investigate the changes of NF-κBp65 and expression of Bcl-2 and Bax relate to apoptosis regulation in brain of spontaneously hypertensive rats( SHRs) and study the protective effect of stellate ganglion block and its mechanism of action. Methods Thirty-two ten-week-old male spontaneously hypertensive rats were divided randomly into four groups: left stellate ganglion block group( group LS),right stellate ganglion block group( group RS),captopril group( group D) and control group( group C). After the experiment,the spontaneously hypertensive rats was intraperitoneally anesthetized with 3% sodium entobarbital( 45 mg / kg) and the brain was quickly removed Bcl-2,bax were investigated by immunohistochemistry and nuclear factor kappa Bp65 was detected by radioimmunoassay. Results Nuclear factor kappa Bp65 of group RS and group D were significantly lower than those of other groups(( P 0. 05). There was no difference in group RS and group D as well as group LS and group C( P 0. 05). Compared with group LS,D and C,SHR brain expression of bcl-2 and bcl-2 / bax in group RS significantly increased( P 0. 05),and bax expression significantly decreased( P 0. 05). Conclusion Right stellate ganglion block has the protective effect on brain of spontaneously hypertensive rats through regulating the expression of NF-κBp65 and apoptosis related genes.
