摘要 目的研究microRNA-34a(mir-34a)在人胃癌细胞中的表达水平及对胃癌SGC-7901细胞增殖、凋亡的影响。方法通过实时定量PCR(RT-PCR)检测人正常胃黏膜上皮细胞GES及人胃癌细胞株AGS、SGC-7901、MKN-45、BGC-823中mir-34a的表达水平。体外将表达人mir-34a(has-mir-34a)慢病毒感染人胃癌SGC-7901细胞。荧光显微镜观察评估细胞感染情况,MTT实验、流式细胞术检测感染后胃癌SGC-7901细胞的增殖、细胞周期及凋亡情况。结果胃癌细胞AGS、SGC-7901、MKN-45、BGC-823中mir-34a的表达水平较正常胃黏膜上皮细胞GES明显降低,其中以胃癌SGC-7901细胞降低最为明显(P<0.01)。与阴性对照组相比,mir-34a感染组SGC-7901细胞增殖明显减慢(P<0.01),G1/G0期细胞比例显著增加(P<0.05),细胞凋亡率显著升高(P<0.01)。结论 mir-34a在多种人胃癌细胞,尤其是SGC-7901细胞中呈低表达。mir-34a可以抑制胃癌SGC7901细胞的增殖,诱导细胞周期停滞及细胞凋亡。 Objective To investigate the expression level of microRNA-34a (miRNA-34a,mir-34a) in human gas-tric cancer cell lines, and explore the role of mir-34a on the proliferation and apoptosis of human gastric cancer cell line SGC-7901. Methods The expression levels of mir-34a in human gastric mucosal epithelial cells GES and hu-man gastric cancer cell lines AGS, SGC-7901, MKN-45, BGC-823 were detected by RT-PCR. Gastric cancer SGC-7901 cells were infected with the lentiviral system expressing mir-34a in vitro. The infected cells status was e-valuated by fluorescence microscope. The proliferation, cell cycle and apoptosis were measured by MTT and flow cytometry. Results Compared with GES, the expression level of mir-34a decreased significantly in gastric cancer cell lines, especially in SGC-7901(P 〈 0. 01). The ratios of proliferation inhibition, cells arrested at G1 / G0 phase and apoptosis were significantly increased in mir-34a lentiviral infected group compared with the negative control group(P 〈 0. 05). Conclusion Mir-34a has lower expression level in some gastric cancer cell lines. Mir-34a can inhibit the proliferation, induce cell cycle arrest and cell apoptosis of gastric cancer cell line SGC7901.
