谷氨酰胺对慢性阻塞性肺疾病患者外周血IL-6及HSP70表达的影响

摘要 目的:观察谷氨酰胺(Gin)对慢性阻塞性肺疾病(COPD)患者外周血单个核细胞(PBMC)中IL-6和热休克蛋白70(HSP70)表达的影响。方法:收集COPD急性加重期(AECOPD)、COPD稳定期(SCOPD)各30例外周血PBMC,将AECOPD、SCOPD患者外周血PBMC各分为Gin组和空白对照组两组,健康组外周血PBMCl5例;应用Real—timePCR法测定五组外周血PBMC中IL-6、HSP70的基因表达。结果:健康对照组较AECOPD和SCOPD空白对照组IL-6、HSP70的表达低,差异有统计学意义(均P〈0.05或P〈001);AECOPD纽和SCOPD组中,用Gin干预的较空白对照组PBMC中HSP70的表达均升高,差异有统计学意义(均P〈0.05),而IL-6表达均下降(均P〈0.05)。结论:Gln可抑制COPD患者炎症因子IL-6的活性,升高HSP70的表达。 Objective: To investigate the effects of glutamine on the expression of IL - 6 and HSP70 in PBMC of patients with COPD . Methods : PBMCs were collected from COPD patients with acute exacerbation (AECOPD group, n = 30) , stable COPD patients (SCOPI) group, n = 30) and 15 health controls. Two COPD groups were further divided into 4 subgroups : blank control and glutamine (Gin) groups. The levels of IL - 6 and HSP70 gene expression in PBMCs were detected by real - time PCR. Results :The mRNA expression of IL - 6 and HSP70 in blank control group of AECOPD and SCOPD were higher than those in the normal group ( P 〈 0. 05 or P 〈 0. 01 ). After treated by glutamine,the expression of HSP70 increased in both AECOPD and SCOPD patients ( P 〈 0. 05 ), while the expression of IL - 6 decreased (P 〈 0. 05 ). Conclusion : Glutamines could inhibit mRNA expression of IL -6 and increase HSP70.
