摘要
目的:探讨天然化合物厚朴酚(Magnolol,Mag)对人乳腺癌细胞转移的抑制作用,并探讨其机制是否与NF-κB信号通路相关。方法:乳腺癌细胞MDA-MB-231经不同浓度Mag处理后,接种到Transwell小室,作用24 h,染色并计穿过Transwell小室的细胞数目;用Western blot法检测不同浓度Mag处理的MDA-MB-231细胞NF-κB信号通路蛋白表达情况。结果:Mag显著抑制乳腺癌细胞的侵袭、迁移和黏附等恶性转移行为,Western blot显示Mag抑制了NF-κB信号通路。结论:Mag具有抑制乳腺癌细胞转移的作用,其机制可能与NF-κB信号通路失活相关。
Objective To explore the effects of natural compound magnolol on metastasis of human breast cancer and investigate whether NF-κB pathway was involved in the effect. Methods MDA-MB-231 cells were treated with magnolol at different concentrations and then cell migration was assessed after 24 h by crystal violet staining and counting. The expression of NF-κB pathway proteins was determined by Western blot analysis. Results Magnolol obviously inhibited the invasion,migration,and adhesion of MDA-MB-231 cells. Western blot demonstrated that magnolol inhibited the NF-κB pathway. Conclusion Magnolol could inhibit the metastasis of human breast cancer MDA-MB-231 cells,which may be relate with the deactivation of NF-κB pathway.
