不同浓度信号分子诱导的小麦TaLr19TLP1基因的表达分析 被引量:2

摘要 为了明确脱落酸(Abscisic acid,ABA)、水杨酸(Salicylic acid,SA)、茉莉酸甲酯(Methyl jasmonate,MeJA)、乙烯(Ethephon,ETH)等信号分子对小麦TaLr19TLP1基因表达的调控作用,在前期研究的基础上,利用半定量RT-PCR对ABA、SA、MeJA和ETH处理后TcLr19小麦中TaLr19TLP1基因表达模式进行分析。结果表明:不同浓度信号分子诱导后不同时间点,TaLr19TLP1基因表达趋势一致,但表达量差异显著。0.5mmol/L ABA、0.5mmol/L SA、0.05mmol/L ETH、0.1mmol/L MeJA诱导后,TaLr19TLP1基因表达高峰出现较早,表达量显著高于相同信号分子其他浓度处理。其中,不同浓度ABA诱导后,TaLr19TLP1基因在不同时间点表达量差异最明显。TaLr19TLP1基因在ETH诱导后6h出现表达高峰,在ABA,MeJA诱导后12h达到表达高峰,在SA诱导后48h出现表达高峰。研究表明,该基因表达受脱落酸、水杨酸、茉莉酸甲酯和乙烯等信号分子的不同程度的诱导,乙烯最先诱导表达,并明确0.5mmol/L ABA、0.5mmol/L SA、0.05mmol/L ETH、0.1mmol/L MeJA为相对最佳诱导浓度。 To study the regulation of different signal molecules,including abscisic acid,salicylic acid,methyl jasmonate and ethylene,on the expression of TaLr19TLP1 gene,the expression profiles of TaLr19TLP1 gene induced by different signal molecules in TcLr19 wheat were detected by semi-quantitative RT-PCR method.The results showed that the expression trend of TaLr19TLP1 gene among different concentration signal molecules was consistent,but expression levels were significantly different.The expression of TaLr19TLP1 gene reached the peak earlier and its expression levels were higher in TcLr19 after treated with 0.5mmol/LABA,0.5mmol/LSA,0.05mmol/L ETH,and 0.1mmol/L MeJA than other concentrations.Especially treated with ABA,the expression levels of TaLr19TLP1 gene had obvious differences at different concentrations.In addition,the accumulation of TaLr19TLP1 gene reached the peak at 6hafter induced by ETH,12 hafter induced by ABA and MeJA,and 48 hafter induced by SA.These results indicated that the expression of TaLr19TLP1 gene can be regulated by these four signal molecules at proper concentrations,and the most firstly induction and expression were found by ETH.The results also indicated that the optimal concentrations were 0.5 mmol/LABA,0.5 mmol/LSA,0.05 mmol/L ETH,and 0.1mmol/L MeJA,respectively.
