大鼠衰老与血浆中T_3、T_4、TSH含量变化相关性随机平行对照研究 被引量:2

摘要 [目的]观察大鼠衰老与血浆中T3、T4、TSH含量变化相关性。[方法]使用随机平行对照方法,将30只大鼠按鼠龄简单随机分成三组,10只/组。将大鼠血浆中T3、T4、TSH检测ELISA试剂盒从4℃冰箱中取出,采用酶联免疫吸附法(ELISA)温静置30min,进行稀释、加样、温育、配液、洗涤、加酶、温育、洗涤、显色处理。每孔加入终止液50μL,终止反应(此时蓝色立即转为黄色)。以空白孔调零,450nm波长下测定各孔的吸光度(OD)值,测定应在加完终止液后15min以内完成。观测血浆T3、T4、TSH含量。[结果]三组T3、T4、TSH、心率有明显差异(P<0.05)。T3指标,6月龄值高于8月龄(P<0.05),8月龄值高于12月龄(P<0.05);T4指标,6月龄值高于8月龄(P<0.05),8月龄值高于12月龄(P<0.05);TSH指标,6月龄值高于8月龄(P<0.05),8月龄值高于12月龄(P<0.05);心率指标,6月龄值高于8月龄和12月龄(P<0.05),而8月龄和12月龄值则没有显著差异(P>0.05)。[结论]SD大鼠月龄增长,SDH活性反应显示能量代谢水平逐渐降低,分泌甲状腺激素的功能亦逐渐降低,大鼠心率随T3、T4、促甲状腺素含量的降低而呈降低的趋势。 [Objective]To observe the plasma of rats with aging the correlation of T3,T4,TSH levels change. [Methods]Using random parallel control method,according to a simple mouse age 30 rats were randomly divided into three groups, 10/group.Will be in rat plasma T3,T4,TSH testing ELISA kit from 4 ℃ refrigerator,by using enzyme-linked immunosorbent(ELISA)WenJing 30 min,dilute, incubate,distribution, liquid, washing,enzym atic,incubate,washing,eolor processing,Each hole to join terminated liquid 50 mu,L termination reaction(blue immediately turned yellow)at this time.With zero blank hole,determine the hole under the 450 nm wavelength absorbance(OD)value,the determination shall be in complete within 15 min after the termination of liquid. Observation of the serum Ta,T4,TSH levels.[Results]The three groups of Ta,T4,TSH,heart rate have obvious difference(P〈0.05).T3 indicators,6 months of age value higher than 8 months of age(P〈0.05),8 months value is higher than 12 months of age(P〈0.05);T4 indicators,6 months of age value higher than 8 months of age(P 〈0.05),8 months value is higher than 12 months of age (P〈0.05);TSH indicators,6 months of age value higher than 8 months of age(P〈0.05),8 months value is higher than 12 months of age(P〈0.05);Heart rate index,6 months of age value higher than 8 months and 12 months of age(P〈0.05),and 8 months and 12 months there is no significant difference(P〉0.05). [Conclusion]SD rats growth,SDH activity reaction shows energy metabolism level gradually reduce, its secretion of thyroid hormone function also gradually reduced.
