氯化钴诱导缺氧对脑胶质瘤T98G细胞株放射敏感性的影响 被引量:1

摘要 [目的]观察氯化钴诱导的缺氧对脑胶质瘤细胞周期的改变及放射敏感性的影响。[方法 ]用氯化钴诱导胶质瘤T98G细胞缺氧,细胞克隆形成实验观察细胞放射敏感性。流式细胞仪检测细胞周期的变化。[结果]T98G细胞在缺氧状态下放射敏感性较常氧下明显降低,差异有统计学意义。氯化钴诱导细胞缺氧后,与常氧状态相比,S期细胞比例由4.7%提高至24.0%,G1期细胞比例由90.4%降至66.7%。[结论]氯化钴诱导的缺氧能改变胶质瘤T98G细胞株的细胞周期,降低细胞的放射敏感性。 [Purpose] To investigate the effect of hypoxia induced by the cobalt chloride(Co Cl2)on the cell cycle and the radiosensitivity of glioma cell lines T98 G. [Methods] Hypoxia of human glioma cell lines T98 G was induced by Co Cl2,and radiosensitivity of cells was detected by colony formation assay. The cell cycle was examined using the flow cytometry. [Results] The radiosensitivity of glioma cell lines T98 G in hypoxia was decreased significantly than that in normoxic state. Results of flow cytometry showed that after treatment by Co Cl2,the percentage of S phase increased from 4.7% to 24.0%; the percentage of G1 phase decreased from 90.4% to 66.7%.[Conclusion] Hypoxia induced by Co Cl2 might change the cell cycle and contribute to the reduction of radiosensitivity of glioma cell lines T98G
