摘要
[目的]探讨冷冻治疗对荷瘤鼠单个核细胞表面程序性死亡配体-1(PD-L1)分子表达的影响。[方法]流式细胞术检测冷冻术后2h、3d、7d对照组和冷冻治疗组荷瘤鼠外周血和脾脏单个核细胞表面PD-L1分子的动态表达。[结果]对照组单个核细胞表面PD-L1表达水平逐渐升高(P<0.01),冷冻治疗组逐渐下降(P<0.05);与对照组相比,冷冻治疗术后2h PDL1分子表达有一过性升高,但3d、7d后显著下降(P<0.01)。[结论 ]单个核细胞表面PD-L1分子表达水平随Lewis肺癌进展逐渐增强,冷冻治疗可能可以通过降低PD-L1分子的表达水平,提高抗肿瘤免疫活性。
[Purpose] To investigate the effect of cryoablation on the expression of programmed death ligand-1(PD-L1) on the surface of mononuclear cells of tumor-bearing mice. [Methods] The expression of PD-L1 on the mononuclear cells of peripheral blood and spleen in control group and cryotherapy group was determined by flow cytometry after 2 hours,3 days,7 days of Argon-Helium cryoablation. [Results] The expression level of PD-L1 on the surface of mononuclear cells gradually increased in the control group(P〈0.01),but it gradually decreased in the cryotherapy group(P 0.05).Compared with control group,the expression level of PD-L1 increased after 2 hours of cryoablation,but significantly decreased after 3 days and 7 days(P〈0.05). [Conclusion] The expression level of PD-L1 on the surface of mononuclear cells increases gradually with tumor progression of Lewis lung cancer,and Argon-Helium cryoablation possibly enhances the anti-tumor immune activity by reducing the expression level of PD-L1.
