铜过载对小鼠肝肾的损伤 被引量:2

摘要 80只32-34 g CD-1小鼠,随机分对照组、铜Ⅰ组、铜Ⅱ组和铜Ⅲ组,铜添加量分别为每千克体重0、2.5、5.0和7.5 mg。Cu Cl2溶解在生理盐水中,腹腔注射8次,每周2次。统计小鼠存活率,火焰原子吸收法检测肝和肾铜含量,全自动生化分析仪检测血清肝、肾功能相关生化参数。常规石蜡切片,HE染色,观察肝和肾病理形态。结果显示:对照组和铜Ⅰ、Ⅱ组无小鼠死亡,铜Ⅲ组存活率仅为20%。铜Ⅱ组和铜Ⅲ组血清谷丙转氨酶、谷草转氨酶活性升高明显,尿素氮和肌酐增加明显,总蛋白和白蛋白轻度减少。铜处理小鼠肝和肾中铜含量增加。肝淤血,肝细胞水肿、变性、坏死;肾淤血,肾小管上皮细胞变性,肾小管阻塞,管腔变小。结果表明,一定量的铜过载可引起小鼠急性死亡,铜在肝和肾沉积,肝、肾形态和功能受损,损伤程度与铜过载量相关。
