摘要
目的观察IL-13对9HTE气道上皮细胞内Egr-1、Bax、Bcl-2 m RNA和蛋白表达的影响,探讨IL-13在气道上皮细胞凋亡中的作用及机制。方法将9HTE气道上皮细胞分为正常对照组、rh IL-13组及rh IL-13+m AIL-13组,用逆转录-多聚酶链反应(RT-PCR)法检测Egr-1、Bax、Bcl-2 m RNA表达水平,用免疫细胞化学(ICC)法检测Egr-1、Bax、Bcl-2蛋白表达水平。结果rh IL-13组Egr-1、Bax m RNA及蛋白表达显著高于正常对照组(P<0.01),Bcl-2 m RNA及蛋白表达显著低于正常对照组(P<0.01);rh IL-13+m AIL-13组Egr-1、Bax m RNA及蛋白表达显著低于rh IL-13组(P<0.01),与正常对照组比较无显著性差异(P>0.05),Bcl-2 m RNA及蛋白表达显著高于rh IL-13组(P<0.01),与正常对照组比较无显著性差异(P>0.05)。结论 IL-13可能通过Egr-1促进9HTE气道上皮细胞凋亡;m AIL-13可以中和IL-13所致的9HTE气道上皮细胞凋亡的作用。
This study aimed to investigate the effects of IL-13 on m RNA and protein expression of Egr-1, Bax and Bcl-2 in 9HTE airway epithelium, and to explore the mechanisms of IL-13 in promoting airway epithelium apoptosis. 9HTE airway epithelium was divided into 3 groups: control group, rh IL-13 group, and rh IL-13 +m AIL-13 group. Egr-1, Bax, and Bcl-2 m RNA were measured with RT-PCR, while Egr-1, Bax, and Bcl-2 protein were measured with ICC. We found that the m RNA and protein levels of Egr-1, Bax in IL-13 group were significantly higher than those in control group(P〈0.01); the m RNA and protein levels of Bcl-2 in IL-13 group were significantly lower than those in control group(P〈0.01); the m RNA and protein levels of Egr-1 and Bax in rh IL-13+ m AIL-13 group were significantly lower than those in rh IL-13 group(P〈0.01), but there was no significant difference as compared with control group(P〈0.05); the m RNA and protein level of Bcl-2 in IL-13 group were significantly higher than those in rh IL-13 group(P〈0.01), but there was no significant difference between rh IL-13+m AIL-13 group and control group(P〈0.05). In conclusion, IL-13 possibly promotes 9HTE airway epithelium apoptosis through Egr-1 pathway; and m AIL-13 may neutralize the effect of IL-13 in promoting apoptosis of 9HTE airway epithelium.
