GSK-3β在胃癌中的表达及其与5-氟尿嘧啶、顺铂药物敏感性之间的关系

Expression of glycogen synthase kinase 3 beta in gastric carcinoma and its relationship with the sensitivity of chemotherapeutic drugs

摘要 目的研究胃癌中糖原合成酶激酶-3β(GSK-3β)蛋白表达及其与5-氟尿嘧啶、顺铂药物敏感性之间的关系。方法收集胃癌新鲜标本50例,提取组织总蛋白,采用蛋白印迹实验(Western blot)检测胃癌及癌旁组织GSK-3β蛋白的表达情况。四甲基噻唑蓝(MTT)法检测不同胃癌组织对5-氟尿嘧啶及顺铂的药物敏感性,分析GSK-3β蛋白的表达与临床病理、药物敏感性之间的关系。结果与癌旁正常组织相比,胃癌组织中GSK-3β的表达显著降低(P<0.05),并且与患者的年龄、分化程度、TNM分期及淋巴结转移有关(P<0.05)。GSK-3β在5-氟尿嘧啶及顺铂耐药病例中的表达量明显低于敏感病例,差异有统计学意义(P<0.05)。结论 GSK-3β的表达下调可能参与了胃癌发生、发展的病理过程,并且与化疗药物耐药性有关。 Objective To explore the expression of glycogen synthase kinase 3 beta(GSK-3β) in gastric carcinoma and its relationship with the clinicopathological features and sensitivity of chemotherapeutic drugs. Methods The expression of GSK-313 in 50 gastric carcinoma and paracancerous specimens was detected by Western blot, and the sensitivities of the tumor to chemotherapeutic drugs were detected by MTF assay. Results GSK-3β protein expression was significantly lower in gastric carcinoma than that in paracan- cerous specimens (P 〈 0.05). The expression of GSK-3 β was correlated with age, differential degree, TNM stage and lymph node me- tastasis (P 〈 0.05 ). GSK-3β protein levels in 5-FU-resistant and cisplatin-resistant cases were lower than those in sensitive cases (P 〈 0. 05 ). Conclusion The low expression of GSK-3β may play an important role in the occurrence and progression of gastric carcino- ma, and is correlated with drug-resistance.
