金天格胶囊对骨质疏松性骨折大鼠血清IL-2、IL-4和TNF-α表达的影响 被引量:14

摘要 目的研究一种新的国家一类中药金天格胶囊对骨质疏松性骨折大鼠骨折愈合过程中细胞因子的影响。方法建立绝经后骨质疏松性骨折大鼠模型,给予雌激素及不同浓度的金天格胶囊灌胃,并于灌胃治疗5周后,采用酶联免疫吸附法(ELISA)测定白细胞介素-2(IL-2)、白细胞介素-4(IL-4)及肿瘤坏死因子-α(TNF-α)的表达。结果模型组大鼠的IL-2、IL-4的阳性表达明显降低、TNF-α的阳性表达明显升高(P<0.05);中、低剂量的金天格胶囊可使大鼠的IL-2、IL-4的阳性表达明显升高、TNF-α的阳性表达明显降低(P<0.05),而高剂量金天格胶囊的干预效果不显著(P>0.05)。结论 1IL-2、IL-4是促进骨质疏松性骨折愈合的细胞因子,与TNF-α的作用相反2金天格胶囊对骨质疏松性骨折大鼠的愈合具有促进作用。 Objective To study the effects of a class of the newstate traditional Chinese medicine,Jintiange capsules,on the expression of cytokines during the fracture healing process in osteoporotic rats. Methods The rat model of postmenopausal osteoporotic fracture was established. Jintiange capsules and different concentrations of estrogen were administered through stomach.After 5 weeks of the treatment,expression of interleukin-2( IL-2),interleukin-4( IL-4),and tumor necrosis factor alpha( TNF-α)were determined using enzyme-linked immunosorbent assay( ELISA). Re sults The positive expression of IL-2 and IL-4decreased significantly,but the expression of TNF- α increased significantly in rats of model group( P〈0. 05). Lowand middle doses of Jintiange capsules increased the positive expression of IL-2 and IL-4,but decreased TNF-α significantly in rat( P〈0. 05).No significant intervention with high dose of Jintiange capsules was found( P〉0. 05). Conclusion 1) IL-2 and IL-4 are cytokines that promote the healing of osteoporotic fractures,which is opposite to the effect of TNF- α. 2) Jintiange capsules can promote the healing of osteoporotic fractures.
