The influence of different modes of oscillation on automatic ELISA test

摘要 目的探讨ELISA试验时,不同振荡混匀方式对全自动酶免系统检测结果的影响。方法选择HBs Ag试剂盒与全自动酶免系统进行匹配试验,根据振荡混匀参数不同,选择低速振荡(B)组和高速振荡(C)组,每组再根据振荡混匀时间(10、30、60 s)不同分为3组(B1-B3、C1-C3),另设手工混匀对照(A)组,每小组采用低值质控血清作为标本进行16孔试验。计算各组均值±标准差(x珋±s)及变异系数(CV),对最终的检测结果进行分析。结果 A组(4.65±0.29)6.2%,B组:B1(3.54±0.46),13.0%;B2(4.08±0.44),10.8%;B3(4.49±0.21),4.7%,C组:C1(3.12±0.71),22.8%;C2(3.86±0.31),8.0%;C3(4.57±0.31)6.8%。各组结果与充分混匀A组比较,只有B3、C3组的结果差异无统计学意义;振荡时间相同的B组和C组比较均无统计学意义。结论作为全自动酶免系统运行的重要环节之一,振荡方式的合理选择会直接影响ELISA结果的准确性。 Objective To investigate the possibility that different modes of oscillation can affect the accuracy of fully automatic ELISA test. Methods According to different parameters of oscillation, low - speed group (B) and high - speed group (C) were selected in which each group was divided into three different groups ( B1 - B3 ,C1 - C3 ) by different blending time (10 s, 30 s, 60 s). In the experiment, the manual group was control group (A). Every group used a low value of serum as control samples to complete the experience in 16 holes. The final test results of each group were analyzed and calculated by x^-±s and coefficient of variation (CV). Results x^-±s and CV in group A were 4.65 ±0.29 and 6.2%, respectively, while those in group B were 3.54 ±0.46 and 13.0% (B1), 4.08 ±0.44 and 10.8% (B2) and 4.49 ±0.21 and4. 7 % ( B3 ), and those in group C were 3.12 ±0.71 and 22.8 % ( C 1 ), 3.86 ±0.31 and 8.0% ( C2), and 4.57 ±0.31 and 6.8% (C3), respectively. In the groups with 10s and 30s blending time, there were statistically significant difference between among groups B, C and control(P 〈 0.05). In the groups with 60s blending time, there was no statistically significant difference between both groups B and C and control (P 〉 0.05). With the same blending time, there were not statistically significant difference between groups B and C(P 〉0.05). Conclusion The oscillation is one of the important link in conducting a fully automatic enzyme system. Reasonable choice of oscillation modes directly affects the accuracy of ELISA experimental results.
