摘要
目的观察分析原发性结肠癌中转录因子Prox-1的表达与淋巴管密度(lymphatic vessel density,LVD)的关系,探讨其临床意义。方法对我院收治的50例结肠癌患者的癌旁非癌组织和正常大肠组织,采用逆转录-聚合酶链反应法检测Prox-1表达情况,采用免疫组化SP法检测对应组织中LVD。结果癌旁非癌组织中Prox-1表达水平显著高于正常组织(t=6.232,P<0.05);癌旁非癌组织LVD显著高于正常组织(t=8.627,P<0.05);癌旁非癌组织Prox-1的表达水平与LVD呈正相关(r=0.621,P<0.05),在浸润深度(t=3.310,P<0.05)、TNM分期(t=3.341,P<0.05)及淋巴转移(t=2.689,P<0.05)上差异有统计学意义。结论结肠癌癌旁非癌组织中Prox-1的表达水平较高,且与淋巴转移、浸润深度及TNM分期有关,可作为结肠癌预后判断的有效预测因子之一。
Objective To explore the expression of the transcription factor Prox-1 in colorectal carcinoma and its relationship with lymphatic vessel density, and clinical significance. Methods Fifty colorectal carcinoma patients were included in this study. RT-PCR was used to detect the expression of Prox-1in paracarcinoma tissues and normal colorectal tissues; immunohistochemical method(SP) was used to examine the lymphatic vessel density(LVD). Results The expression of Prox-1 m R NA in paracarcinoma tissues was signif icantly higher than that in nor mal tissues(t = 6.232, P〈0.05); the LV D in paracarcinoma tissues was signif icantly higher than that in nor mal tissues(t =8.627, P〈0.05). The expression of Prox-1 m R NA in paracarcinoma tissues had positively linear relation with LV D(r = 0.621, P〈0.05). The expression of Prox-1 of paracarcinoma tissues in dimension of invasion depth(t =3.310, P〈0.05),T N M staging(t =3.341, P〈0.05) and ly mph node metastasis(t =2.689, P〈0.05) showed signif icant difference. Conclusion The overexpression of Prox-1 i n pa raca rci noma t issue is related to ly mphat ic met ast asis, depth of i nvasion and T N M st age. T he expression of Prox-1 can be one of impor tant prognostic factors for colon cancer.
