山稔子油敷剂对深Ⅱ度烧伤小鼠TNF-α、ICAM-1的影响 被引量:2

Effect of Sanrenzi Oil Dressing on TNF-α and ICAM-1 of Deep Second-degree Burn Mice

摘要 目的观察山稔子油敷剂对深Ⅱ度烧伤小鼠的疗效及其对肿瘤坏死因子-α(TNF-α)、细胞间粘附因子1(ICAM-1)的影响。方法将30只小鼠随机分成正常组,模型组和山稔子油敷剂组,正常组6只,其余两组各12只。造模4 h后给药干预,观察烧伤的一般指标。小鼠分两批处死,4 d时,模型组和山稔子油敷剂组随机处死6只,12 d后再处死余下的小鼠,眼球采血,分离烧伤组织,HE染色观察皮肤组织的病理变化,酶联免疫(Elisa)、免疫组化(IHC)检测烧伤组织TNF-α,ICAM-1的表达。结果 4 d和12 d山稔子油敷剂组创面积明显小于相对应模型组的面积(P<0.05,P<0.01),炎症细胞浸润减少,12 d个别小鼠创面全部愈合。4 d和12 d时山稔子油敷剂组ICAM-1表达均低于模型组(P<0.05),4 d时山稔子油敷剂组TNF-α表达低于模型组(P<0.05),12 d时山稔子油敷剂组明显高于模型组(P<0.05)。结论山稔子油敷剂具有抗炎、减少疤痕、缩短愈合时间的作用,急性期减少TNF-α,修复期维持其相对量的表达量。急性期、修复期均减少ICAM-1的表达。山稔子油敷剂对小鼠深Ⅱ度烧伤的疗效可能与TNF-α、ICAM-1有关。 Objective To investigate the therapeutic effects of Sanrenzi(SRZ) Oil dressing and its influence on tumor necrosis factor-α (TNF-α) and intercellular adhesion molecule 1 (ICAM-1) expression in deep second-degree burn mice. Methods Thirty mice were divided into normal control group(n=6), model group(n=12) and SRZ Oil dressing group(n=12). Treatment was carried out 4 hours after the modeling, and then we observed the general data of burning in each group. On treatment day 4, 6 mice were sampled separately from the model group and SRZ Oil dressing group for sacrifice, and one treatment day 12, the left mice in the two groups were also executed for the separation of burn tissue. The pathological changes in the burn skin was observed after HE staining, and the expression levels of TNF- and ICAM-1 in the burning tissue were detected by enzyme-linked immunosorbent assay (ELISA) and immunohistochemistry (IHC). Results Compared with the model group, the lesion area of SRZ Oil dressing group became significantly smaller on treatment day 4 and 12 (P 〈 0.05, P 〈 0.01 ), the infiltration of the inflammatory cells was significantly reduced, and the lesion was completely cured in some mice of SRZ Oil dressing group on treatment day 12. The expression level of ICAM-1 in SRZ Oil dressing group was significantly lower than that of the model group on treatment day 4 and 12(P 〈 0.05), TNF-α expression level in SRZ Oil dressing group was significantly lower than that of the model group on treatment day 4(P 〈 0.05), but became higher on treatment day 12(P 〈 0.05). Conclusion SRZ Oil dressing has obvious effects on inhibiting inflammation, reducing scar, and shortening cure timein mice with deep second-degree burning, and its therapeutic mechanism is probably related to the regulation of TNF-α and ICAM-1 expression.
