HPLC法同时测定救必应药材中4种成分的含量

Simultaneous Determination of Four Constituents in Cortex Ilicis Rotundae with HPLC

目的建立同时测定救必应药材中紫丁香苷、丁香脂素4’-0书-D-吡喃葡萄糖苷、长梗冬青苷、铁冬青酸含量的方法。方法采用Kromasil100—5C18色谱柱（250mm×4．6mm，5μm），流动相为乙腈（A）-0．02％三氟乙酸水溶液（B），梯度洗脱，流速为1．0mL·min-1，检测波长为210nm，柱温为30℃。结果4种成分均达到基线分离，线性关系良好（r≥0．9999）；平均加样回收率为98．16％。101．3％，RSD〈3．0％（n=6）。结论该方法简单准确可靠，可为救必应药材的质量控制提供科学依据。

Objective To establish a HPLC method for simultaneous determination of syringin, syringaresinol 4＇-0- β-D-glucopyranoside, peduncloside and rotundic acid in Cortex Ilicis Rotundae. Methods The analysis was performed on a Kromasil 100-5 Cls column （250 mm×4.6 mm, 5μm） with the mobile phase consisting of acetonitrile（A）-0.02 % trifluoroacetic acid （B） by gradient elution at a flow rate of 1.0 mL≥min-1. The detection wavelength was set at 210 nm, and the column temperature was set at 30℃. Results Baseline separation was obtained and linear relationship was good （r≥0.9 999） for the four constituents. The average recoveries were 98.16 %-101.3 % and RSD 〈 3.0 % （n=6）. Conclusion The established method is rapid and reliable, which could provide scientific evidence for the quality control of Cortex Ilicis Rotundae.
