摘要
目的探讨侧脑室注射醒脑静对脑出血(ICH)大鼠血肿周围TLR4及炎性因子TNF-α、IL-1β表达的影响。方法采用大鼠尾状核自体血注射法制作大鼠ICH模型,随机分成模型组、侧脑室组、静脉组,并设立正常组。对各组大鼠进行行为学评分,采用HE染色观察炎性浸润情况,通过免疫组化检测TLR4、TNF-α和IL-1β的表达。结果侧脑室组第24、48、72小时的神经功能评分显著低于静脉组,差异有统计学意义(P<0.05)。侧脑室组的炎性细胞浸润较静脉组轻。模型组的TLR4、IL-1β和TNF-α平均积分光密度值显著高于静脉组和侧脑室组,差异有统计学意义(P<0.05)。侧脑室组的TLR4和TNF-α平均积分光密度值显著低于静脉组,差异有统计学意义(P<0.05)。结论侧脑室注射醒脑静能显著抑制ICH血肿周围的炎性细胞浸润及TLR4、TNF-α表达。
Objective To explore the influence of intracerebroventricular injection of Xingnaojing on expression of TLR4 and inflammatory factor such as TNF-α,IL-1β in the cerebral hemorrhage (ICH). Methods The model of ICH was maked by injected autoblood from tail artery into right caudate nucleus in rats,and which were randomly divided into the model group,the lateral ventricle group,the venous group and the normal group.Behavior scores of rats in each group was observed.Inflammatory infiltration was observed through HE stain.Immunohistochemistry was used to detect the expression of TLR4,TNF-α and IL-1β. Results The score of the neurological function in the lateral ventricle group at the 24^th,48^th,72^th hours was lower than that in vein group,with significant difference(P〈0.05).The infiltration of inflammatory cells in the lateral ventricle group was lower than that in the venous group.The average integral optical density of TLR4,IL-1βand TNF-α in the modeI group was higher than that in the venous group and the lateral ventricle group,with significant difference(P〈0.05).The average integral optical density of TLR4 and TNF-α in the lateral ventricle group was lower than that that in the venous group,with significant difference(P〈0.05). Conclusion Intracerebroventricular injection of Xingnaojing can significantly inhibit the infiltration of inflammatory cells and expression of TLR4, TNF-α of ICH around the hematoma.
