灯盏花素对糖尿病肾病大鼠肾组织肾小管超微结构的影响 被引量:4

Effect of breviscapine on renal tubule ultrastructure in streptozotocin(STZ)- induced diabetic nephropathy rats

摘要 目的观察灯盏花素对链脲佐菌素(STZ)诱导糖尿病肾病(DN)大鼠肾组织肾小管超微结构的影响。方法STZ诱导糖尿病肾病SD大鼠模型,雌性SD大鼠随机分为正常对照组(C)、DN模型组(DN)、DN+灯盏花素治疗组(DS)三组。模型建立后,于第12周末,测量血糖(BG)、胆固醇(CHO)、低密度脂蛋白胆固醇(LDL-C)、尿素氮(BUN)、血肌酐(Scr)、尿白蛋白排泄率(UAER)。处死大鼠后,电镜下观察糖尿病大鼠的肾组织肾小管超微结构改变。结果实验12周末,与DN组比较,DS组大鼠CHO、LDL-C、尿素氮(BUN)、血肌酐(Scr)、UAER均降低,分别降低了11%、22%、50%、29%、45%(P<0.05);C组肾小管上皮细胞大小一致,形态正常;DN组可见肾小管上皮细胞重度水肿,小管间可见大量胶原纤维;DS组肾小管上皮细胞病变程度较DN组轻。结论灯盏花素具有保护糖尿病肾组织肾小管功能,从而减缓糖尿病肾病的发生和发展。 Objective To investigate the effects of breviscapine on renal tubule ultrastructure of rat with streptozotocin(STZ)-induced diabetic nephropathy(DN). Method The STZ –induced diabetic SD female rats were divided into normalcontrol group(C),diabetic nephropathy group(DN),diabetic breviscapine treatment group(DS). The parameters of blood glu-cose(BG), cholesterol(CHO),low density lipoprotein cholesterol(LDL-C),urea nitrogen(BUN),serum creatinine(Scr),urinary albumin excretion rate(UAER)in rats of each groups were measured after 12 weeks. Changes in renal tissue of renaltubule ultrastructure in diabetic rats were observed under electron microscope. Results After 12 weeks the levels of CHO,LDL-C,BUN,Scr and UAER were significantly reduced compared with that of DN group,(P〈0.05),The size of renal tubularepithelial cells in control group was uniform with normal morphology; renal tubular epithelial cells in DN group showed severeedema, tubule showed a large amount of collagen fibers in group DS; renal tubular epithelial cell lesions was less than that inDN group. Conclusion Breviscapine can protect the renal tubular function in diabetic rats retarding the occurrence and de-velopment of diabetic nephropathy.
