摘要 目的:探讨响应面法优化哈蟆油肌苷提取的最佳工艺条件。方法:在单因素实验的基础上,采用响应面分析法对影响加热提取哈蟆油核苷类成分肌苷得率的主要因素(甲醇体积分数、提取时间和料液比)进行优化,建立影响因素与肌苷之间的函数关系。结果:获得最佳工艺条件为:甲醇体积分数为0,即为纯水提取,时间为67.0 min,料液比(g∶ml)为1∶71,在此最佳条件下,得到肌苷的含量为23.31μg/g,试验结果与模型预测值相符。结论:该研究可为哈蟆油的深入开发和广泛应用提供科学依据。 OBJECTIVE: To optimize surface methodology. METHODS: Based the extraction technology of Inosine from Oviductus Ranae by response on the single factor experiment, the main factors (volume fraction of methanol, extraction time and solid-liquid ratio) associated with yield of inosine (extracted by heating extration) from Oviductus Ranae were optimzied by response surface methodology. The functional relationship between the influencing factors and the inosine yield was established. RESULTS: The optimal extraction conditions of Inosine from Oviductus Ranae were achieved when methanol volume fraction was 0, i. e. , the extraeting solvent was water, with extracting time of 67.0 min and solid-liquid ratio of 1: 71. Under these conditions, the maximal yield of Inosine was 23.31μg/g, which was consistent with the predicted values. CONCLUSIONS: This study provides a scientific basis for further development wand extensive use of Oviductus Ranae.
