摘要
目的:探讨肿瘤坏死因子-α(TNF-α)对骨髓间充质干细胞(BMSCs)凋亡的影响。方法:分离培养大鼠BMSCs,经成骨、成脂诱导分化鉴定后,分别用浓度为0、20、40μg/L的TNF-α对其进行刺激。24 h后,通过细胞活性检测观察不同浓度TNF-α对BMSCs的活性影响;Tunel染色观察不同浓度TNF-α对BMSCs凋亡的影响;Western blot检测不同浓度TNF-α刺激后,BMSCs中Bax、Bcl-2蛋白的表达量。结果:BMSCs经体外培养扩增后,成骨、成脂鉴定均为阳性。台盼蓝染色、Tunel染色结果显示,随TNF-α浓度增高,BMSCs的死亡率增高,细胞凋亡率则呈浓度依赖性增高(P<0.05);Western blot结果显示,随TNF-α浓度增高,BMSCs中Bax表达逐渐增加,Bcl-2表达逐渐降低(P<0.05)。结论:TNF-α可诱导BMSCs的凋亡。
AIM: To investigate the effects of tumor necrosis factor- α( TNF-α) on the apoptosis of rat bone marrow mesenchymal stem cells( BMSCs). METHODS: Mouse BMSCs were cultured and stimulated with TNF-α( μg / L) at 0,20 and 40 respectively for 24 h. Cell activity staining and Tunel staining were used to determine the apoptosis of BMSCs. The expression of bax and Bcl-2 protein was examined by Western blot. RESULUTS: TNF- αdose dependently decreased the survival and increased the apoptosis of BMSCs. TNF- α up- regulated Bax expression and down- regulated bcl-2 expression( P〈0. 05). CONCLUSION: TNF-α can induce the apoptosis of BMSCs.
