益气活血方动员骨髓干细胞对心肌IRI大鼠血管新生的影响

Effects of Yiqi Huoxue Decoction on Myocardial IRI Rat Angiogenesis with Bone Marrow Stem Cells Mobilization

目的:研究益气活血方对心肌缺血再灌注损伤(IRI)鼠心肌细胞CD34+和血管内皮细胞生长因子(VEGF)蛋白表达的影响,以探讨其对骨髓干细胞动员作用及血管新生作用机制。方法:大鼠120只,随机分为假手术组,IRI组;rh EPO动员组;益气活血方组;联合动员组,每组24只,制备心肌IRI模型后,各组再随机分为7d,14 d,28 d三个时间段亚组,每组8只。采用免疫组化法检测各组CD34+及VEGF蛋白表达。结果:与模型组比较,rh EPO动员组和益气活血方组CD34+蛋白表达和VEGF蛋白表达明显升高,差异具有统计学意义(P﹤0.05,P﹤0.01);与rh EPO动员组和益气活血方组比较,联合动员组CD34+蛋白表达和VEGF蛋白表达明显升高,差异具有统计学意义(P﹤0.05,P﹤0.01)。结论:益气活血方具有动员骨髓干细胞作用,益气活血方联合动员剂对骨髓干细胞动员和血管新生具有增强促进作用。

Objective：To investigate the effects of Yiqi Huoxue Decoction on myocardial ischemia- reperfusion injury （IRI） rat cardiomyocytes CD34 ＋ and vascular endothelial growth factor（VEGF） protein expression and to further ex- plore the mechanisms of bone marrow stem cell mobilization and angiogenesis. Methods ：120 rats were randomly divided in- to sham group（ n = 24）, IRI group （ n = 24）, rhEPO mobilization group（ n = 24）, Yiqi Huoxue group （ n = 24）, combined mobilization group（ n = 24 ）. After myocardial IRI model preparation, each group was randomly divided into 7 d （n = 8 ）, 14 d（n =8） ,28 d （n =8） three time periods subgroups. Detect each groupg CD3d ＋ and VEGF protein expressions by immunohistochemistry. Compared with the model group,the CD34 ＋ and VEGF protein expressions of rhEPO mobilization group and Yiqi Huoxue group were significantly increased （P 〈 0.05, P 〈 0.01 ）. Compared with rhEPO mobilization group and Yiqi Huoxue group, the CD34 ＋ and VEGF protein expressions of combined mobilization group was significantly increased（P 〈0.05 ,P 〈 0.01 ）. Conclusion： Yiqi Huoxue Decoction has the function of bone marrow stem cell mobiliza- tion. And Yiqi Huoxue combined mobilization formula plays an enhanced role in bone marrow stem cell mobilization and angiogenesis.
