摘要
目的:观察心痛方对急性心肌缺血大鼠血浆中CD40/CD40L、PS/PSGL-1表达及大鼠心肌梗死面积的影响。方法:40只SD大鼠,随机分成假手术组、模型组、心痛方组、欣康对照组,采用结扎法制备心肌缺血模型,NBT染色法测定心肌梗死面积,酶联免疫法检测血浆中CD40/CD40L、PS/PSGL-1的表达。结果:与模型组相比,心痛方和欣康均缩小心肌梗死面积(P<0.01),且心痛方组较欣康组更有优势(P<0.01)。心痛方组、欣康组及模型组造模后分别与假手术组相比,血浆中CD40/CD40L、PS/PSGL-1均明显增高(P<0.01);用药后心痛方组、欣康组分别与模型组相比,CD40/CD40L、PS/PSGL-1的表达均明显降低(P<0.05);心痛方与欣康在抑制CD40/CD40L、PS/PSGL-1表达方面作用相近。结论:心痛方能抑制急性心肌缺血大鼠血浆CD40/CD40L及PS/PSGL-1的表达、缩小大鼠心肌梗死面积。
Objective : To observe the effect of Xintong Decoction on expressions of CD40/CD40L and PS/PSGL - 1 in plasma and myocardial infarction size of rats. Methods: 40 SD rats were randomly divided into sham operation group, model group, Xintong Decoction group, Xinkang control group. To establish the myocardial infarction model by coronary artery ligation method,the myocardial infarct size was determined by NBT staining. The imrnunohistochemistry was used to detect the expressions of CD40/CD40L and PS/PSGL- 1 in the plasma. Results: Compared with the model group,the Xintong Decoction group and Xinkang group can reduce myocardial infarction size of rats ( P 〈 0.01 ), and Xintong Decoc- tion group was better than Xinkang group (P 〈0.01 ). When Xintong Decoction group, Xinkang group and model group compared with the sham operation group after modeling, respectively, CD40/CD40L and PS/PSGL - 1 in the plasma were all significantly increased (P 〈 0.01 ) in three groups. When Xintong Decoction group and Xinkang group compared with the model group after drugs use, CD40/CD40L and PS/PSGL - 1 were significantly lower ( P 〈 0.05 ) in Xintong Decoction group and Xinkang group. Xintong Decoction group and Xinkang group had the similar inhibitory effect on CD40/CD40L and PS/PSGL - 1 expressions. Conclusion : Xintong Decoction could reduce the myocardial infarction area of rats and inhibit CD40/CD40L and PS/PSGL - 1 expressions in the plasma of acute myocardial ischemia rats.
