摘要
目的:研究三叶青乙醇提取物不同洗脱组分的抗氧化活性差异。方法:三叶青用70%乙醇回流提取得到三叶青乙醇提取物,上D-101大孔树脂,分别用不同浓度的乙醇进行洗脱,得到不同洗脱物TDG-W、TDG-30%、TDG-50%、TDG-70%、TDG-90%。比较不同洗脱物的总黄酮、总多酚含量并采用DPPH法、FRAP法和ORAC法测定其抗氧化活性,2,6-二叔丁基-4-甲基苯酚(BHT)作为阳性对照。结果:TDG-W、TDG-30%、TDG-50%、TDG-70%、TDG-90%中总黄酮和总多酚含量分别为2.69%、33.25%、61.12%、40.54%、4.12%和0.62%、3.48%、5.07%、4.35%、2.70%。其清除DPPH自由基能力、还原能力和总抗氧化能力均随总黄酮和总多酚浓度的增加而上升;5种洗脱物的抗氧化能力与所含总黄酮和总多酚的量有一定相关性。结论:三叶青不同洗脱组分均有较强的抗氧化能力,对体外自由基有较好的清除作用。
Objective:To investigate the antioxidant activity of different elution fractions from Tetrastigmahemsleyanum Diels et. Gilg ethanol extracts in vitro. Methods : Five elution fractions ( TDG - W, TDG - 30%, TDG - 50%, TDG - 70% and TDG -90% )isolated and purified from Tetrastigmahemsleyanum Diels et. Gilg ethanol extracts using D - 101 macro- porous resins were valuated for the antioxidant activities by DPPH radical scavenging, reducing power and antioxidant activities. In addition, the results were compared with natural and synthetic antioxidants called BHT. The levels of total phenolics and total flavonoids of the elution fractions were also determined. Results : The contents of total flavonoids and total phenolics of the different elution fractions from Tetrastigmahemsleyanum Diels et. Gilg ethanol extracts were 2.69%, 33.25% ,61.12% ,40.54% ,4.12% and 0.62%, 3.48% ,5.07% ,4.35% and 2.70%, respectively. The scavenging capacity of DPPH, reducing power and antioxidant activities increased with the increasing concentration of total flavonoids and total phenolics. The antioxidant capacity of five elution fractions was somewhat associated with the contents of total fla- vonoids and total phenolics. Conclusion:From the results it can be concluded that the different elution fractions from Tet- rastigmahemsleyanum Diels et. Gilg ethanol extracts have strong antioxidant capacity to have free radical scavenged in vitro.
