4种中药复方对DN大鼠超微组织结构的影响 被引量:5

Impact of Four Chinese Herbal Compounds on Glomerular Glycoprotein Deposition and Ultrastructure of Diabetic Nephropathy in Rats

摘要 目的:观察4种中药复方对糖尿病肾病(DN,diabetic nephropathy)大鼠肾小球的组织超微结构、肾小球足细胞的影响。方法:取链脲霉素诱导的SD大鼠DN模型72只,分成6组,模型对照组、益气养阴方(参芪地黄汤20 g·kg-1)组、滋补肝肾方(杞菊地黄汤20 g·kg-1)组、补气养血方(当归补血汤20 g·kg-1)组、温肾健脾方(附子理中丸20 g·kg-1)组和阳性对照(缬沙坦25 mg·kg-1)组,每组12只;另取12只大鼠为正常对照组。连续经口灌胃给药8周后,进行肾脏组织HE染色病理切片和电镜肾脏病理形态学分析。结果:各给药组大鼠肾脏重量、肾重/体质量比值均有一定的降低趋势(P>0.05),肾小球截面积和肾小球体积均明显降低(P<0.05,P<0.01),滋补肝肾组、补气养血组和温肾健脾组大鼠肾小球囊腔面积显著降低(P<0.05,P<0.01),各组肾小球囊腔面积/肾小球截面积比值均有降低的趋势(P>0.05),肾小球基底膜厚度与模型对照组比有所减轻,足细胞足突融合、变平的现象与模型对照组比有所改善。结论:4种中药复方能不同程度的降低糖尿病大鼠肾小球截面积、肾小球体积、囊腔面积和囊腔面积/肾小球截面积,改善糖尿病大鼠肾小球足细胞的融合和变平现象,减少基底膜增厚程度。 Objective: To observe the impact of four Chinese herbal compounds on glomerular deposition glycoprotein and organic uhrastructure of diabetic nephropathy in rats. Methods:Take seventy- two STZ -induced diabetic nephropatl, rats and divide them into model control group, supplementing Qi and nourishing yin group (Shenqi Dihuang Decoc- tion,20 g/kg), nourishing liver and kidney group (Qiju Dihuang Decoction, 20 g/kg), supplementing Qi and nourishing blood group (Danggui Buxue Decoction,20 g/kg,wanning kidney and invigorating spleen group (Fuzi Lizhong Pill ,20 g/ kg) and positive control group (valsartan 25 mg/kg). Each group had 12 rats,taking another 12 normal rats for the nor- mal control. Administration drug for eight weeks, HE and PAS staining for the kidney tissue and electron microscope for slices and observe kidney pathological morphology. Results:Each groups had certain reduce trend in the rat kidney weight and kidney weight/body weight ratio ( P 〉 0.05 ), and significantly lower glomerulus sectional area and volume ( P 〈 0.05 ,P 〈0.01 ). Nourishing liver and kidney group, supplementing Qi and nourishing blood group and warming kidney and invigorating spleen group can significantly reduce bowman's capsule area (P 〈 0.05 ,P 〈 0.01 ). Each groups all had reduce trend of area ratio of bowman's capsule and glomerulus ( P 〉 0.05) and significantly reduced glomerular PAS posi- tive area and the total optical density(P 〈0.01 ). Moreover,it can alleviate glomerular basement membrane thickness and improve electron dense deposit and the phenomenon of podocyte foot process fusion and flatten compared with those of the model control group. Conclusion:Four kinds of Chinese herbal compounds can inhibit glomerulus glycoprotein deposition, glomerular area, glomentlar volume, area ratio of bowman's capsule and glomerulus in different degrees and improve glomerular podocyte fusion and flatten phenomenon and reduce the degree of thickening of basement membrane in diabetic rats.
