钙敏感受体在大鼠扩张型心肌病心肌损伤中的作用

摘要 目的观察阿霉素(doxorubicin,DOX)诱导大鼠扩张型心肌病(dilated cardiomyopathy,DCM)钙敏感受体(calcium-sensing receptor,Ca SR)表达的变化,探讨钙敏感受体在扩张型心肌病中的作用。方法 40只雄性Wistar大鼠,随机分为对照组(n=10)和模型组(n=30),模型组大鼠分为3周组、6周组和9周组。采用腹腔注射阿霉素(2.5mg·kg-1·w-1)建立大鼠DCM模型。应用超声心动图检测各组大鼠左心室内径以及心功能。HE染色观察大鼠心肌超微结构变化。Western blot和免疫组化法检测心肌组织Ca SR蛋白的表达情况。结果 (1)与对照组相比,9周组大鼠左室舒张末期内径(LVDD)明显增加(P<0.01),左室射血分数(LVEF)明显降低,;(2)随着病程延长模型组大鼠心肌Ca SR表达明显降低(P<0.01)。结论在大鼠扩张型心肌病的发生过程中,Ca SR表达逐渐降低,可能导致钙稳态紊乱参与DCM的发生。 Objective observation of doxorubicin (doxorubicin, DOX) in rats induced by dilated cardiomyopathy (dilated cardiomyopathy, DCM) calcium -sensing receptor (calcium -sensing receptor, CaSR) the expression change of calcium -sensing receptor in the function of dilated cardiomyopathy. Methods Forty male Wistar rats were randomly divided into two groups ( n = 10) and the model group ( n =30), models of rats were divided into 3 weeks, 6 weeks and 9 weeks group. By intraperitoneal injection of ADRIAMYCIN (2.5mg·kg^-1·w^-1 ) establishing rat model of DCM. Echocardiography in detecting left ventricular diameter as well as cardiac function in each group. HE staining observation of changes of myocardial uhrastructure in rats. Western blot and Immunohistochemistry assay for detection of myocardial tissue expression of CaSR. Results compared with the control group, 9 - week rats left ventricular end - diastolic diameter (LVDD) were significantly increased (p 〈 0. 01 ), left ventricular ejection fraction (LVEF) was significantly reduced, second as duration model of rat myocardial CaSR expression significantly reduced (P 〈 0.01 ). Findings in rats with dilated cardiomyopathy during CaSR expression decreases, which might lead to disturbance of Calcium Homeostasis in participating in DCM. Enter text you want to translate!
