基因修饰的骨髓间充质干细胞移植对大鼠急性心肌梗死的疗效观察

摘要 目的:探究基因修饰的骨髓间充质干细胞移植对大鼠模型急性心肌梗死的治疗作用。方法:选取15 d龄SD乳鼠20只,平均分为观察组(血管内皮生长因子组)和对照组(DMEM组)两组,每组各10只大鼠,制备急性心急梗死模型后,观察组大鼠行基因修饰的骨髓间充质干细胞移植,对照组大鼠注射无血清DMEM,移植1个月后,观察两组大鼠的心脏功能情况。结果:观察组与对照组大鼠LVEF分别为(73.45±9.17)%、(43.26±6.33)%,观察组明显高于对照组,两组比较差异有统计学意义(P<0.05);两组大鼠镜下均见左室瘢痕组织形成,有新生血管生成,并出现左室重塑特征,观察组大鼠心脏改变较对照组明显;观察组大鼠与对照组大鼠血管密度分别为(45.69±4.28)、(17.41±3.67),观察组明显高于对照组,两组比较差异有统计学意义(P<0.05)。 Objective: To explore the curative effect observe of gene modified bone marrow mesenchymal stem cells transplantation on acute myocardial infarction in rats.Method: 20 SD rats of 15 day old were selected and divided into two groups, the observation group(vascular endothelial growth factor group) and control group(DMEM group), 10 rats in each group, the preparation of acute myocardial infarction model, the observation group rats underwent gene modified bone marrow mesenchymal stem cells transplantation,the control group rats were injected with serum free DMEM, after one month transplantation,the cardiac function of two groups were observed. Result: The observation group and the control group LVEF rats were respectively(73.45±9.17)% and(43.26±6.33)%, the observation group was significantly higher than that in the control group, the difference was statistically significant(P〈0.05);two groups of rats were found with microscope in the formation of left ventricular scar tissue,with angiogenesis and the emergence of remodeling left ventricular, compared with the control group,the observation group rats heart changed significantly; the observation group and the control group of rats with vascular density of rats were respectively(45.69±4.28) and(17.41±3.67), the observation group was significantly higher than that in the control group, the difference was statistically significant(P〈0.05). Conclusion: Vascular endothelial growth factor gene modified bone marrow mesenchymal stem cells transplantation has good therapeutic effect on acute myocardial ischemia in rats.
