摘要 目的运用代谢组学及生物效应网络方法研究阿魏酸生物效应的机制。方法采用高效液相色谱-质谱(HPLC-MS)联用技术测定阿魏酸给药组与空白组大鼠血清,质谱数据采用MATLAB软件处理,利用偏最小二乘辨别法分析空白组、阿魏酸给药组之间的的代谢差异,并通过查询Ch EBI、KEGG等数据库确定经变量重要性投影所选取的潜在生物标志物的化学结构。结果阿魏酸给药组与空白组比较有显著的代谢差异,与其相关的两种代谢物及其作用靶点和代谢途径分别为二磷酸腺苷(ADP)、三磷酸腺苷(ATP)以及脆性组氨酸三联基因(FHIT)蛋白和ATP敏感性钾通道(KATP)。结论代谢组学及生物效应网络方法能用于阿魏酸生物效应机制的研究,为进一步揭示药物作用机制提供了新方法。 Objective To study the biological effects mechanism of ferulic acid by metabolomic method and biomolecular network. Methods HPLC-MS was used to analyze the serum metabolome in ferulic acid administration group and control group rats,MS data were processed by MATLAB software. With multivariate statistical analysis of serum metabolite profiles,a clear separation among ferulic acid administration group and control group was achieved,and the structure of potential biomarker was selected according to the parameters of variable importance in the projection( VIP) confirmed by Ch EBI、KEGG databases. Results The difference of metabolism between ferulic acid group and control group was significantly. Two compounds,related action targets and metabolic pathways were confirmed,which were adenosine diphosphate( ADP),adenosine triphosphate( ATP),fragile histidine triad( FHIT) gene protein and ATP sensitive potassium channel( KATP),respectively. Conclusion Metabolomics and biomolecular network can be used to investigate the biological effects mechanism of quercetin,which provid a new method to further revealing mechanism of drug action.
