摘要
研究了枯草芽孢杆菌(Bacillus subtilis)GS8菌株液体发酵条件,为提高其活性及次级代谢产物的产量提供技术支持。通过单因子试验和正交试验,对该菌株的最适发酵培养基成分及发酵条件进行了优化。结果显示:枯草芽孢杆菌GS8菌株培养基组分最佳配比为蔗糖2%、牛肉浸膏0.8%、氯化铵0.2%、硫酸镁0.02%,最佳发酵条件为培养时间4 d、培养温度27℃、初始p H 7.0、摇床转速150 r/min、250 m L装液量100m L、接菌量为装液量的2%。在最佳发酵培养基和培养条件下,GS8菌株发酵滤液对葡萄炭疽病菌的抑制率提高了13.1%。
The research was to investigate the optimal conditions for liquid fermentation of Bacillus subtilis GS8 strain in order to provide technical support for increasing production of its secondary metabolites. The single factor experiments and orthogonal experiments were adopted to optimize the liquid fermentation medium and conditions. The results showed that the optimal compositions of cultural medium were as follows: sucrose 2%,beef extract 0. 8%,NH4 Cl 0. 2%,Mg SO40. 02%. The optimal fermentation conditions were as follows: fermentation time 4 d,cultured temperature 27 ℃,initial p H 7. 0,rotating speed 150 r / min,medium capacity 100 m L /250 m L,and inoculation rate2%. Inhibitory rate of strain GS8 fermentation filtrate against Colletotrichum gloeosporiodes increased by 13. 1% under the optimal fermentation medium and culture conditions.
